A 66‐year‐old male with osteoclast‐like giant cell tumor of the liver that arose in the non‐cirrhotlc liver is presented. The liver tests were almost normal, and plasma levels of alpha‐fetoprotein and carcinoembryonic antigen were within normal limits. The findings of liver scan by 99m Tc phytate, celiac angiography, and CT scans are described for the first time for this rare neoplasm, showing a large, unresectable liver tumor. Histologically, the tumor mainly consisted of osteoclast‐like giant cells and mononuclear cells, which were focally arranged in a vaguely trabecular pattern and sarcomatous pattern. By an electromicroscopic study, however, no definitive evidence was obtained whether it arose from epithelial cells or nonepithellal cells. Various clinicopathological features were described and compared with previously reported cases including two cases arising in the liver.

OSTEOCLAST‐LIKE GIANT CELL TUMOR OF THE LIVER

Lymph nodes from 21 cases of malignant lymphoma of a centrocytic (mantle cell) type, (ML, cc (mc)) were examined. All the cases had monoclonal surface immunoglobulin (sig) M and/or D, but were negative for CD 10 (CALLA), and CD11c (LeuM5). Lymphoma cells with CD25 (anti‐Tac)+, CD5 (Leu1)+, and alkaline phosphatase (ALPase) ‐ in eight cases showed bone marrow involvement (10–66% of the nucleated cells; mean 32± 18%) but with no leukemic changes. These eight cases has a similar phenotype and were distributed by the lymphoma cells to the examined B‐chronic lymphocytic leukemia. Seven cases showed an infiltration of CD25 ‐ , CD5+, and AL Pase ‐ lymphoma cells, in which only two cases showed focal bone marrow involvement.There was a close relationship between CD25 expression and bone marrow invasion by the lymphoma cells in ML, cc(mc). Three of the six CD25 ‐ and CD5 ‐ cases presented zonal proliferation of AlPase+ lymphoma cells with round nuciee and a high anti‐proliferating cell nuclear antigen/cyclin (PCNA/c) rate in the mantle zone and paracortex, accompanied by a prominent interdigitating dendritic and histiocytic cell reaction. Examined CD25 ‐ , CD5 ‐ and ALPase+ lymphoma showed a neoplastic counterpart of so‐called marginal zone lymphocytes, which was different from other cases of ML, cc (mc). Lymphoma cells in ML, cc(mc), except for those of the so‐called marginal zone lymphoma, might be derived from slgM+, D+/ ‐ , CD25+/ ‐ , CD5+ /‐ , ALPase ‐ , CD10 ‐ and CD11c‐ lymphocytes present in the mantle zone and primary lymph follicles. Other types of lowgrade B cell neoplasms that were examined had different tumor cell markersfor ig, CD5, CD10, CD11c, a higher PCNA/c rate, and prominent dendritic cells and histiocytic reaction when compared with those of ML, cc (mc).

Clinicopathologic, enzyme and hisotchemical studies of centrocytic (mantle cell) lymphoma: Comparison with other types of low‐grade B cell lymphoma bases on the updated Kiel classification

Senile Nagoya, Shibata, Yasuda (NSY) mice developed amyloidosis and died from renal failure as a result of amyloidosis. NSY mice were first reported as experimental congenital diabetic mice by Shibata et al . in 1980. This study questioned whether NSY mice died from diabetic nephropathy. The authors of the present study investigated the life span and cause of death in these micde. The life span of NSY mice was found to be 618.7 ± 72.5 days. NSY mice that lived for more than 400 days showed rising blood uread nitrogen and large amounts of amyloid deposits in the glomerulus of the kidneys. NSY mice died of renal amyloidosis. Immunological methods revealed that AApoAll was evident in the amyloid deposits of NSY mice. Apart from the kidneys, amyloid deposition was also found in the tongue, esojphagus, stomach, small intestine, large intestine, rectum, lung, heart and adrenal glands. Amyloid deposits were found to a slight degree In the liver and the spleen. The most dominant amyloid deposition in NSY mice was seen in the glomerulus of the kidneys. From the point of view of amyloid depositional distribution, NSY mice were unique compared with other spontaneous amyloid mice.

Spontaneous amyloidosis in senile NSY mice

The appearance of orthochromatic erythroblasts in the peripheral blood was evaluated as an index of the initial stage of leukemia induced in rats by the oral administration of N,N'-2,7-fluorenylenebisacetamide, 1-ethyl-1-nitrosourea and 1-butyl-1-nitrosourea. Solitary leukemia lesions found mainly in the bone marrow and composed of one or a few nodular foci were found by using erythroblasts as an index. The time interval from the first appearance of erythroblasts to autopsy was short in solitary lesions, slightly longer in scattered lesions, and even longer in diffuse leukemia lesions. Although the appearance of orthochromatic erythroblasts in the peripheral blood was due to several causes other than leukemia, this appearance was useful as an index of the initial stage of chemically induced leukemia in rats.

A new human T cell line with a chromosomal abnormality (47,XY,+2), designated AS-IIA, was established by coculturing peripheral blood leukocytes of a healthy adult male with a lethally irradiated human T lymphotropic virus type II (HTLV-II)-infected simian leukocyte cell line (Si-IIA). A polymerase chain reaction method showed that this interleukin-2 (IL-2)-dependent cell line possessed the HTLV-II provirus genome; the cells also reacted with HTLV-II-positive human sera, anti-HTLV-I/II p24, and anti-HTLV-II gp46 antibodies. AS-IIA cells expressed the suppressor/cytotoxic T cell markers CD3+, CD4-, CD8+, CD25+, and HLA-DR+, with later conversion to CD8-. These cells showed better proliferation than other human HTLV-II-infected cell lines with normal karyotypes, but were not transplantable into severe combined immunodeficiency mice. Virus production from AS-IIA was confirmed not only by electron microscopic examination, which revealed mature and immature type C virus particles, but also by the capacity of the line to immortalize human T cells. These results suggest that HTLV-II shows broad tropism for T cells including CD4+ or CD8+, and that not only Si-IIA, but also AS-IIA, are good sources of HTLV-II. The authors of the present study believe that AS-IIA may be a useful human T cell line for the investigation of HTLV-II in comparison with HTLV-I.

Learn more.Copy URL Share a linkShare onEmailFacebookTwitterLinkedInRedditWechat Summary A modified indirect Coons' immunofluorescent technic was used to determine whether or not there exist circulating anti-liver tissue antibodies in the sera of 20 patients with various chronic liver diseases and 25 patients other than liver diseases. The normal liver tissue obtained from victims of traffic accident within 3 hours of death was used as antigen. For the control gamma-globulin in the normal liver was stained by direct Goons' technic. Even in the normal liver, intracytoplasmic gamma-globulin was observed in a certain cytoplasm of the hepatic cells in three out of five cases, and also in the cytoplasm of Kupffer's cells in one of them. The incidence of circulating antibody to the cytoplasm of hepatic cell was 13 out of 20 cases (65%), anti-nucleus antibody was one in 20 (5%), and anti-cytoplasm of bile ductular cell antibody was two in 20 patients (10%) with chronic liver diseases, whereas the sera of normal persons as well as of patients other than liver diseases were all proved to be negative. The anti-nucleus antibody was non-specific in species and organs. No close correlation was found among the appearance of anti-liver tissue antibodies, gamma-globulin quantities in sera, and tissue reactions such as single cell necrosis, piecemeal necrosis, and cell infiltration in the Glisson's capsule. Nonetheless, the appearance of circulating antibody to the cytoplasm of hepatic cell was detected in seven of 8 patients (87.5%) with active chronic hepatitis. The results imply an etiological role of anti-liver tissue antibody on the progression and prolongation of liver diseases. AN IMMUNOFLUORESCENT STUDY OF CIRCULATING ANTI-LIVER ANTIBODIES IN CHRONIC LIVER DISEASES

In order to study the possible biological differences between anaplastic and typical seminoma, the following factors were studied in 11 cases of anaplastic seminoma and 15 cases of typical seminoma: mitotic activity, proliferating cell nuclear antigen (PCNA) expression, immunohistochemical analyses for cytokeratin, vimentin, placental alkaline phosphatase (PLAP), β‐human chorionic gonadotropin (β‐hCG), a‐fetoprotein (AFP) and c‐myc oncoprotein. Anaplastic seminoma was classified according to Mostofi's criteria, which is primarily based on the mitotic activity of the tumor. Mitotic activity was evaluated by both mitotic count and rate. Statistically significant correlations were observed between mitotic count and mitotic rate ( R = 0.891), and between the mitotic count and PCNA labeling index ( R = 0.792), in both typical and anaplastic seminomas. Immunostaining patterns for cytokeratin, vimentin, PLAP, β‐hCG, AFP and c‐myc oncoprotein were not significantly different between typical and anaplastic seminoma. The present data indicated that no apparent clinicopathologic and immunohistochemical parameters discerning anaplastic seminoma from typical seminoma were present, when identifying anaplastic seminoma on the basis of high mitotic count. Anaplastic seminoma may therefore simply represent seminoma with high proliferative activity.

Immunohistochemical comparison between anaplastic seminoma and typical seminoma

Elastic fibers in 15 blebs and 17 bullae with spontaneous pneumothorax were studied by means of electron microscopy and light and electron microscopic immunohistochemistry for elastin and alpha 1-antitrypsin. Blebs were formed in association with focal organized alveoli, and bullae were formed in association with pulmonary emphysema. Both blebs and bullae had abnormal elastic fibers. Ultrastructurally, abnormal elastic fibers of blebs and bullae consisted of accumulated thick and fine fibers. Accumulated thick elastic fibers showed vacuolar changes and electron-dense granular deposits, and they were associated with spiraling collagen fibrils. These thick elastic fibers reacted evenly with antielastin antibody and also reacted with anti-alpha 1-antitrypsin antibody. They are thought to be degraded elastic fibers. Accumulated fine elastic fibers consisted of bundles of microfibrils and granular amorphous components, and they reacted with anti-elastin and anti-alpha 1-antitrypsin antibody. These fine elastic fibers are thought to be not only newly formed in the process of organization but also degraded. It is suggested that elastic fibers of blebs and bullae are degraded due to an imbalance between elastase and alpha 1-antitrypsin.

Histogenesis of abnormal elastic fibers in blebs and bullae of patients with spontaneous pneumothorax: Ultrastructural and immunohistochemical studies

A case of primary carcinoid tumor arising in the liver of a 69 year old woman with no endocrine symptoms is reported. Histopathologically, the tumor was diagnosed initially as a hepatocellular carcinoma in the biopsy specimen, and was shown subsequently to be a carcinoid tumor, demonstrating diffuse positive staining with Grimelius method. Mucin stained with periodic acid‐Schiff (PAS), alcian‐blue, and mucicar‐mine, and was shown partially in the glandular structures. Immunohistochemically, most of the tumor cells stained positively for chromogranin‐A, epithelial membrane antigen (EMA) and neuron specific enolase (NSE). Ultrastructural examination revealed electron‐dense core granules, measuring 40‐120 nm in diameter in some of the tumor cells. Intensive and careful searches pre‐ and post‐operatively revealed no other primary source of tumor other than the liver. The patient was reported well with no symptoms 3 1 / 2 years after the operation.

Primary hepatic carcinoid tumor

Laminin, type IV collagen and fibronectin were examined immunohistochemically in the invasive component of breast carcinomas. Laminin was expressed around the invasive carcinoma cell nests in 38 (54%) of 71 cases. Immunoreactivity for type IV collagen was observed around the invasive carcinoma cell nests or the stroma apart from carcinoma cells in 44 (80%) of 55 cases. Fibronectin was strongly expressed in the stroma only in 75 (99%) of 76 cases. The expression of laminin significantly correlated with tubular formation in the invasive carcinoma cell nests and showed a tendency to be correlative to estrogen receptor (ER) and progesterone receptor (PgR) of carcinoma tissue, but no correlation among laminin expression, histological type, the age of patients, tumor size and lymph node metastasis was noted. Type IV collagen and fibronectin did not correlate to any clinicopathological factors such as histological type, grade of differentiation, the age of patients, tumor size, lymph node metastasis, ER and PgR status. No concordant expression of these extracellular matrices was seen.
