A racemic mixture of a new bicyclononane aldehyde, (1RS,5SR,6RS)-5-hydroxybicyclo[4.3.0]non-2-ene-2-carbaldehyde (1) was isolated from the fruits of Amomum tsao-ko, together with 12 known compounds (2–13). The structure of 1 was determined on the basis of extensive spectroscopic analysis, including 1D and 2D NMR data. The antiproliferative activity of compounds 1–13 was assessed in the murine neuroblastoma cell line N2a.

Bicyclononane Aldehydes and Antiproliferative Constituents fromAmomum tsao-ko

Evaluation of the topical anti-inflammatory activity of chloroform and ethyl acetate extracts of RUMEX NEPALENSIS roots in a TPA-induced acute inflammation mouse model demonstrated a significant reduction in ear edema. The extracts were further tested on purified enzymes for COX-1 and COX-2 inhibition to elucidate their mechanism of action, and a strong inhibition was observed. Six anthraquinones and two naphthalene derivatives were isolated from the ethyl acetate extract. Among the isolated compounds, emodin was found to be a potent inhibitor with slight selectivity towards COX-2, and nepodin exhibited selectivity towards COX-1. Emodin, endocrocin, and nepodin also exhibited significant topical anti-inflammatory activity in mice. Interestingly, nepodin showed better radical scavenging activity than trolox and ascorbic acid against DPPH and ABTS radicals. The strong radical scavenging activity of chloroform and ethyl acetate extracts could be explained by the presence of nepodin as well as by the high phenolic content of the ethyl acetate extract. Thus, the anti-inflammatory effect of R. NEPALENSIS roots was assumed to be mediated through COX inhibition by anthraquinones and naphthalene derivatives and through the radical scavenging activities of naphthalene derivatives.

Anti-inflammatory, Cyclooxygenase (COX)-2, COX-1 Inhibitory, and Free Radical Scavenging Effects ofRumex nepalensis

Abstract Glucose homeostasis is maintained by antagonistic hormones such as insulin and glucagon as well as by regulation of glucose absorption, gluconeogenesis, biosynthesis and mobilization of glycogen, glucose consumption in all tissues and glomerular filtration, and reabsorption of glucose in the kidneys. Glucose enters or leaves cells mainly with the help of two membrane integrated transporters belonging either to the family of facilitative glucose transporters (GLUTs) or to the family of sodium glucose cotransporters (SGLTs). The intestinal glucose absorption by endothelial cells is managed by SGLT1, the transfer from them to the blood by GLUT2. In the kidney SGLT2 and SGLT1 are responsible for reabsorption of filtered glucose from the primary urine, and GLUT2 and GLUT1 enable the transport of glucose from epithelial cells back into the blood stream. The flavonoid phlorizin was isolated from the bark of apple trees and shown to cause glucosuria. Phlorizin is an inhibitor of SGLT1 and SGLT2. With phlorizin as lead compound, specific inhibitors of SGLT2 were developed in the last decade and some of them have been approved for treatment mainly of type 2 diabetes. Inhibition of SGLT2 eliminates excess glucose via the urine. In recent times, the dual SGLT1/SGLT2 inhibitory activity of phlorizin has served as a model for the development and testing of new drugs exhibiting both activities. Besides phlorizin, also some other flavonoids and especially flavonoid enriched plant extracts have been investigated for their potency to reduce postprandial blood glucose levels which can be helpful in the prevention and supplementary treatment especially of type 2 diabetes.

Four diterpenes with biological activity were isolated from Salvia deserta roots. Taxodione was considered leishmanicidal with an IC50 value of 1.46 µM (0.46 mg/L) against Leishmania donovani and also exhibited antifungal and antimicrobial activities. Ferruginol displayed the greatest activity [24-h IC50 of 4.5 µM (1.29 mg/L)] against the fish pathogenic bacteria Streptococcus iniae. The crude extract fraction that contained the isolated compounds 7-O-acetylhorminone and horminone showed stronger in vitro antibacterial activity (1.3 mg/L for Staphylococcus aureus and 1.1 mg/L for methicillin-resistant S. aureus) than the compounds tested alone. 7-O-Acetylhorminone and horminone exhibited a synergistic effect against methicillin-resistant S. aureus (FIC of 0.2), and horminone had better activity against S. aureus with respect to other compounds isolated from S. deserta roots. In larvicidal bioassays, these extracts and isolated pure compounds did not show any activity at the highest dose of 125 mg/L against 1-d-old Aedes aegypti larvae.

Antimicrobial and Antileishmanial Activities of Diterpenoids Isolated from the Roots of Salvia deserta

Significant health and socio-economic challenges are posed by renal diseases, leading to millions of deaths annually. The costs associated with treating and caring for patients with renal diseases are considerable. Current therapies rely on synthetic drugs that often come with side effects. However, phytoestrogens, natural compounds, are emerging as promising renal protective agents. They offer a relatively safe, effective, and cost-efficient alternative to existing therapies. Phytoestrogens being structurally similarity to 17‐β‐estradiol, bind to estrogen receptors and produces both beneficial and, in some cases, harmful health effects. Sirtuins activation has been shown promising in mitigating fibrosis and inflammation in renal tissues. Specifically, SIRT1, which is a crucial regulator of metabolic activities, plays a role in protecting against nephrotoxicity, reducing albuminuria, safeguarding podocytes, and lowering reactive oxygen species in diabetic glomerular injury. Numerous studies have highlighted phytoestrogens ability to activate sirtuins, strengthening antioxidant defense, promoting mitochondrial biogenesis, and playing a vital role in renal protection during kidney injury. These findings support further investigation into the potential role of phytoestrogens in renal protection. This manuscript reviews the potential of phytoestrogens such as resveratrol, genistein, coumesterol, daidzein, and formononetin in regulating sirtuin activity, particularly SIRT1, and thereby providing renal protection. Understanding these mechanisms is crucial for designing effective treatment strategies using naturally occurring phytochemicals against renal diseases.

Phytoestrogens and Sirtuin Activation for Renal Protection: A Review of Potential Therapeutic Strategies

Alopecia is a common dermatological disorder of patchy hair loss with substantial patient burden. Phytotherapeutic compounds are increasingly used as a source of new therapeutic options. This review aimed to synthesize the evidence on plant species in hair growth and the methodological aspects of in vivo experimental models. The systematic scoping review was conducted following the PRISMA checklist, Joanna Briggs Institute and in accordance with Cochrane. A systematic search was carried out in the Pubmed, Scopus, Web of Science, and SciELO databases. In vivo experiments that evaluated hair growth activity using natural substances of plant origin were included. Data collection and analysis: A total of 1,250 studies were identified, of which 175 were included for qualitative synthesis. Of these, 128 used mice, 37 rats, 10 rabbits, 1 guinea pig, and 1 sheep as animal models. The methodologies mapped were: hair growth analysis, histological analysis, immunohistochemistry, gene expression analysis, Western blot, enzyme-linked immunosorbent assay, and biochemical analysis. Minoxidil and finasteride were the most commonly used positive controls. The studies evaluated plant species (166), algae (11) or isolated substances (31). Overall 152 plant species and 37 isolated substances were identified. This is the first systematic scoping review on methodological aspects of in vivo hair growth activity. We created a checklist to be completed by authors to allow data comparison and reproducibility, facilitate data interpretation by readers and ensure better quality of evidence. This work may become a valuable tool for future research and contribute to significant advances in hair growth studies.

Astragalus adsurgens, a significant forage plant cultivated in arid regions of northwest China, remains underexplored for its triterpenoid saponins and medicinal properties compared to the extensively studied Astragalus membranaceus. To explore the phytochemical profile of A. adsurgens for its potential application in the medical field, we employed ultra-pressure liquid chromatography coupled with tandem mass spectrometry-based method to identify cycloartane-type triterpenes. Eventually, five new cycloartane-type triterpenoids, adsurgosides A‒D (1‒4) and 3-methyl-3,4-seco-cyclostellanol (5), together with two known analogues cycloastragenol (6) and cyclopycanthogenin (7), were isolated from the roots of A. adsurgens. Their structures were elucidated using 1D and 2D NMR analyses in combination with HRESIMS data. Additionally, a comparative study on the distribution patterns of these compounds revealed qualitative and quantitative variations between A. adsurgens and A. membranaceus. Our findings not only identified an alternative plant for isolating cycloartane-type triterpenoids but also offer new insights into the chemical properties of A. adsurgens.

Phytochemical characterization and comparative analysis of cycloartane-type triterpenes in Astragalus adsurgens and Astragalus membranaceus

Abstract Aloe vera is commonly used as traditional medicine for cutaneous wound healing. Nonetheless, the wound healing mechanisms of Aloe vera remain unclear. This review aims to provide insight into the molecular mechanisms of Aloe vera in promoting cutaneous wound healing, with particular emphasis on the mechanisms that stimulate cell proliferation and migration. Aloe vera has been shown to upregulate growth factors such as keratinocyte growth factor-1 (KGF-1), transforming growth factor-β (TGF-β), cyclin D1, insulin-like growth factor 1 (IGF-1), vascular endothelial growth factor (VEGF), basic fibroblastic growth factor (bFGF), and microfibril-associated glycoprotein 4 (MFAP4), as well as collagen, fibrillin, elastin, α-smooth muscle actin (α-SMA), integrins, and platelet endothelial cell adhesion molecule 1 (PECAM-1, also known as CD31), while downregulating the expression of matrix metalloproteinases (MMPs). In addition, Aloe vera was also found to upregulate PI3K/Akt and MAPK pathways, as well as the TGF-β signalling pathway via Smad proteins. Furthermore, molecular docking studies revealed that certain chemical constituents of Aloe vera bind to some of the forementioned growth factors or signalling molecules. With regards to current applications, although human clinical trials have reported positive results from using Aloe vera in healing open wounds and burns and alleviating some inflammatory skin diseases, the current commercial uses of Aloe vera remain largely focused on cosmetic products. Thus, greater advances are required to promote the use of Aloe vera products in clinical settings.
