The ability of particle-bound and fluid-phase ligands (IgG, C3b, and ConA) to induce metabolic activation in cytochalasin B-treated human neutrophils was studied. Different activation mechanisms of the neutrophils are proposed, since the activation of cytochalasin B-treated neutrophils is not dependent on whether or not the ligands are particle-bound but not on the nature of ligand per se.

The chemotactic responsiveness of granulocytes obtained from patients with ulcerative colitis was compared to granulocytes from healthy individuals, using the LMAT (Leukocyte Migration Under Agarose Technique). A depression of chemotaxis found in ulcerative colitis patients was seen to be related to the disease activity. This reduction in granulocyte migration could not be corrected by pre-incubation of cell suspensions with tinidazole. The pattern observed for granulocyte chemotaxis in ulcerative colitis was discussed with reference to use as an aid in the differential diagnosis of ulcerative colitis and Crohn's disease.

GRANULOCYTE CHEMOTAXIS IN ULCERATIVE COLITIS

The distribution of T lymphocyte subsets was determined in 22 patients with endometrial carcinoma stage I. Following surgery and postoperative radiotherapy the patients had been randomized® into two groups: group A patients received no further treatment, whereas group B patients were treated with 17 a-hydroxyprogesterone caproate for one year. T cells and T cell subsets were estimated by an indirect immunofluorescence technique using the monoclonal antibodies UCHT3 (mature T cells), OKT4 (T helper cells), and OKT8 (T suppressor/cytotoxic cells). When examined 3–12 months after randomization the two groups had similar numbers of UCHT3+ cells and OKT4+ cells. Group B patients, however, had a significantly lower proportion of OKT8+ cells and hence a significantly higher OKT4/OKT8 ratio than group A patients. It is concluded that progestogen therapy has some immunomodulating effects.

A human serum was resolved by DEAE cellulose chromatography and immunosorbence to isolate the serum component involved in the precipitation. Testing of the final product against antisera to various serum proteins and analysis by SDS-PAGE revealed only serum transferrin. This preparation and transferrin obtained commercially produced precipitation in agar gel against the gonococcal extract. The bacterium was agglutinated by transferrin in solution or when carried by latex particles, and particles sensitized with the bacterial extract were agglutinated by transferrin. Human blood lymphocytes were stimulated in vitro by four Staphylococcus aureus strains. Activation of immunoglobulin-secreting cells was determined by a reverse plaque forming cell (PFC) assay, and proliferation by quantitation of thymidine incorporation. Whole killed S. aureus were slightly more efficient than water-soluble preparations in the form of sonicated extracts and culture supernatants. Two S. aureus strains rich in protein A (Cowan I and E 2371) and one S. aureus strain deficient in protein A (E 1369) were potent B-lymphocyte stimulators inducing maximal activity on day 6 of culture. Another S. aureus strain deficient in protein A (Wood 46) did not possess++ human lymphocyte stimulating capacity.
