Abstract The influence of several process parameters (starter, sourdough refreshment, dough yield, ingredients) on sourdough fermentation was investigated. This was realised by online measurement of both pH and gas pressure in sourdough in a closed air‐tight reactor. Firstly, the effect of sourdough refreshment on both gas pressure and acidification was evaluated and shown to be accompanied by an increase in both acidification and gas pressure evolution rates. Secondly, two commercial starters (BRS and BIO) differing in their microflora were compared as regards to their gassing power and acidifying properties. BRS exhibited faster acidification but less gassing power. Moreover, the variation of sourdough yield for BRS starter induced a different evolution of the acidification profile. Sourdough fermentation varied also depending on the presence of NaCl. 2% of NaCl induced a lower gassing power in sourdough by inhibiting the yeasts.

Influence of Several Process Parameters on Sourdough Fermentation

Abstract The degradation undergone by grape cluster stems (woody component of vine bagasse), an agroindustrial waste, was investigated during the semi‐solid‐state cultivation of Phanerochaete chrysosporium BKM‐F‐1767 (ATCC 24725). For this, the content of lignin, cellulose and hemicellulose in grape cluster stems was determined before and after the enzymatic process. It was found that about 20% of Klason lignin, 48% of hemicellulose and 5% of cellulose were degraded during the process, being the ligninolytic enzymes (manganese‐dependent peroxidase and lignin peroxidase) produced by such cultures responsible for the degradation of grape cluster stems. In parallel, semi‐solid‐state cultures of P. chrysosporium grown on an inert support (cubes of nylon sponge), which is not susceptible to undergoing degradation during the enzymatic process, were used as reference cultures. In addition, the in vivo decolourisation of a model dye, the polymeric dye Poly R‐478, by both grape cluster stem and nylon cultures was studied in order to assess their degradative ability. A percentage of biological decolourisation higher than 90% after four days of dye addition was obtained using nylon sponge cultures, whereas grape cluster stem cultures led to a decolourisation of around 70% after eight days of dye incubation. The lower percentage of dye degradation achieved by the cultures grown on grape cluster stems was due to the enzymes produced, which were not only employed in the decolourisation of the dye but also in the degradation of the support, as indicated by the data mentioned above.

Biodegradation of Grape Cluster Stems and Ligninolytic Enzyme Production by Phanerochaete chrysosporium during Semi‐Solid‐State Cultivation

Abstract While phospholipid fatty acid (PLFA) profiling is a well‐established method used for the determination of bacterial and eukaryotic organisms in soil ecology, phospholipid etherlipid (PLEL) analyses for the characterisation of Archaea is a rather new approach. Analyses of PLEL derived isoprenoid side chains by GC/MS provided a broad picture of the archaeal community in a mixed soil extract, as lipids previously identified in isolates belonging to the kingdoms Eury‐ and Crenarchaeota were covered. Furthermore, ether‐linked isoprenoid hydrocarbons, which have not been detected in archaeal isolates and monomethyl‐branched alkanes which have only been found in hyperthermophilic bacteria, were detected in these soil extracts. Monomethyl‐branched alkanes were the most dominant ones and accounted for 43.4% of the total identified ether‐linked hydrocarbons, followed by straight chain (unbranched) and isoprenoid hydrocarbons, which accounted for 34.6 and 15.5%, respectively.

Abstract This paper deals with the ozonation and biological treatment for achieving the mineralisation of the non‐biodegradable compound naphthalene‐1,5‐disulphonic acid (NADSA; C 10 H 8 O 6 S 2 ), a basic chemical for dyes and building materials. This combination of ozonation and biological treatment is expected to minimise the ozone consumption compared to ozonation alone. To examine the influence of ozone on the biodegradability of NADSA, a lab‐scale plant was run semi‐continuously and batch tests were conducted for comparison. For total ozonation, 1.8 mol O 3 /mol DOC have to be used. For optimum biodegradation, only 0.8 mol O 3 /mol DOC are needed. The results show that by using a combination of ozonation and biological treatment the ozone consumption can be reduced by more than 50% compared to ozonation alone. Some of the intermediate products are isolated and suggestions for their chemical structures are made. A proposal for the first steps of ozonation is also presented to illustrate a possible pathway of ozonation. All the major identifiable compounds are non‐biodegradable. From these results, it is quite clear that further intermediate products are formed, which are biodegradable.
