Objective To translate the English version of fear of coronavirus disease 2019(COVID-19) scale(FCV-19 S) into Chinese and assess its reliability and validity. Methods FCV-19 S was translated and culture-adapted to form a Chinese version of fear of COVID-19 scale(FCV-19 S-C). A total of 334 questionnaires were sent out online, including FCV-19 S-C, Chinese version of depression, anxiety and stress scale(DASS-C21), impact of event scale-revised(IES-R) and public stigma scale. The reliability and validity of FCV-19 S-C and the influencing factors of COVID-19 fear were analyzed with the survey data. Results FCV-19 S-C contained 7 items. One common factor was extracted by exploratory factor analysis, reflecting that all items in the scale belonged to the common factor, which could explain 69.5% of total variation. The load of item factors ranged from 0.780 to 0.873, showing good construct validity. The total score of FCV-19 S-C was positively correlated with the total scores of DASS-C21 and IES-R(r=0.403 and 0.471, both P<0.01), indicating that the scale had good concurrent validity. The Cronbach’s α coefficient of FCV-19 S-C was 0.924, showing good reliability. Linear regression analysis showed that the influences of COVID-19 on the psychological level and family income could predict the total score of FCV-19 S-C(β=0.62 and 0.20, both P<0.01). The total score of FCV-19 S-C could predict the total score of the public stigma scale(β=0.37, P<0.01). Conclusion FCV-19 S-C has good reliability and validity, and can be used as a tool to understand the public fear of COVID-19 in China. The fear of COVID-19 is related to the loss of family income and the public stigma of COVID-19 patients.

Objective To analyze the diagnosis and treatment data after the adjustment of various measures in the hospital fever clinic during the coronavirus disease 2019 (COVID-19) outbreak, so as to provide experience for the prevention and control of COVID-19 in non-epidemic areas Methods We collected the diagnosis and treatment data from the fever clinic of Changhai Hospital of Naval Medical University (Second Military Medical University) in non-epidemic area from Dec 21, 2019 to Feb 22, 2020 and Jan 1 to Mar 5, 2019, and divided them into four groups: Group A (during the 2020 COVID-19 epidemic), from Jan 22 to Feb 22, 2020;group B (before the COVID-19 epidemic), from Dec 21, 2019 to Jan 21, 2020;group C (in the same period as group A in 2019), from Feb 2 to Mar 5, 2019;and group D (in the same period as group B in 2019), from Jan 1 to Feb 1, 2019 Year-on-year and month-on-month comparisons of the above data were conducted Results The overall situation of the fever clinic around the Spring Festival in 2020 was similar to that of the same period in 2019 The year-on-year comparison results showed that during the COVID-19 epidemic, the number of daily patients in the fever clinic was significantly decreased ([114 3±62 9] cases vs [171 7±37 0] cases), the single consultation time of each physician was prolonged ([11 7±1 8] min vs [6 5±1 3] min), but the average waiting time of patients was decreased ([7 5±0 6] min vs [22 8±1 5] min) (all P<0 05) The month-on-month comparison results indicated that the number of daily patients in the fever clinic was significantly decreased during the COVID-19 epidemic ([114 3±62 9] cases vs [216 3±41 8] cases), the single consultation time of each physician was prolonged [( 11 7±1 8] min vs[ 5 6±0 8] min), but the average waiting time of patients was decreased ([7 5±0 6] min vs [23 3±3 2] min) (all P<0 05) The proportion of computed tomography (CT) examination during the 2020 COVID-19 epidemic was significantly higher than that in the same period of 2019 (7 9%[ 292/3 658] vs 3 7% [206/5 493]) Although the examination time was prolonged due to strengthened protective measures ([12 5±3 8] min vs [10 0±3 2] min) (P<0 05), the time for patients to have the CT examination report was significantly shortened ([10 6±2 5] min vs [58 4±9 6] min) (P<0 01) Conclusion During the COVID-19 epidemic, measures such as adjusting the relevant procedures of the fever clinic and equipping special CT machine in non-epidemic area can reduce the daily consultation time of physicians, improving the diagnosis and treatment efficiency and reducing cross infection

Fever clinic in non-epidemic area during coronavirus disease 2019 outbreak: An analysis of countermeasures

Objective To isolate the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) from nasal/throat swabs of coronavirus disease 19 (COVID-19) patients Methods Three nasal/throat swab samples from COVID-19 patients in Shanghai were treated with TPCK trypsin and were used to treat Vero E6 cells inoculated in 96-well plates When most of the cells showed obvious cytopathy, the cell culture supernatants were collected We then detected the viral nucleic acid by fluorescent quantitative real-time polymerase chain reaction, and amplified the gene fragment of the virus receptor binding domain (RBD) by reverse transcription polymerase chain reaction After amplification and culture, the virus was used to infect the Vero E6 cells inoculated in 96-well plates The cytopathy was observed and the virus protein was detected by immunofluorescence Results The Vero E6 cells that cultured with two of three nasal/pharyngeal swab samples showed obvious cytopathic effect and newly synthesized viral nucleic acid was detected in the supernatants of the cell culture The amplified RBD sequence was completely consistent with the corresponding fragment of SARS-CoV-2 isolated earlier Virus-infected Vero E6 cells showed cytopathies rapidly and could react with the monoclonal antibody against nucleocapsid protein (N protein) and spike protein (S protein) of SARS-CoV-2, and convalescence sera of COVID-19 patients Conclusion Two SARS-CoV-2 strains were successfully isolated from two nasal/throat swab samples of COVID-19 patients in Shanghai, which provides evidence for the mechanism research on the infection and pathogenesis of SARS-CoV-2 as well as the development of drugs and vaccines against SARS-CoV-2

Isolation and identification of two strains of severe acute respiratory syndrome coronavirus 2 from coronavirus disease 2019 patients in Shanghai

Objective To investigate the efficacy of neutralizing antibodies induced by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) receptor-binding domain (RBD) and spike (S) protein S1 subunit Methods The SARS-CoV-2 RBD and mouse immunoglobulin G1 (IgG1) Fc fragment (mFc) fusion protein expression plasmid pVRCRBD- mFc was constructed and transfected into human embryonic kidney 293T cells The RBD-mFc fusion protein in the cell supernatants was detected by Western blotting The effect of RBD-mFc in cell supernatants and CHO recombinant S1-human IgG1 Fc (S1-hFc) fusion protein on SARS-CoV-2 infection was detected by microneutralization test BALB/c mice were immunized with plasmid pVRC-RBD-mFc and S1-hFc fusion protein via intramuscular injection Anti-S1 IgG antibodies in mouse sera were detected by enzyme-linked immunosorbent assay (ELISA), and the virus neutralization activity of mouse sera was detected by microneutralization test Results The RBD-mFc fusion protein could be detected in the culture supernatants of 293T cells transfected with the plasmid pVRC-RBD-mFc, the concentrated supernatants and the S1- hFc fusion protein could inhibit SARS-CoV-2 infection on Vero E6 cells in a concentration-dependent manner Anti-S1 IgG antibodies could be detected in the sera of mice immunized with plasmid pVRC-RBD-mFc and S1-hFc fusion protein, and the sera of both groups could neutralize SARS-CoV-2 infection The serum antibody titers and virus neutralization activity of S1- hFc fusion protein immunized mice were significantly higher than those of plasmid pVRC-RBD-mFc immunized mice (both P<0 01) Conclusion Both SARS-CoV-2 RBD and S1 subunit may be used as effective vaccine antigens Compared with DNA vaccine, recombinant subunit vaccine can induce neutralizing antibody more effectively

Objective To explore the characteristics and influencing factors of fatigue in officers and soldiers during the closed management of coronavirus disease 2019 (COVID-19), so as to provide evidence for developing specific strategies on psychological assistance in major public health emergencies. Methods The fatigue status and stress level were evaluated by multidimensional fatigue inventory, military personnel psychological self-evaluation test and self-designed epidemic effect questionnaire in 199 officers and soldiers from an army unit, and the relevant data were analyzed by SPSS 21.0 softwase. Results The total fatigue score was 47.96±12.52, with power loss (50.25%) as the main manifestation, and the stress rate was 7.54% (15/199). There were significant differences in the total fatigue scores concerning gender, job title, service age, educational background, number of closed days, and the impact of the epidemic on themselves (all P<0.05). The total fatigue score and the 3 factors (physical fatigue, mental fatigue and power loss) were positively correlated with the stress level (P<0.05 or P<0.01). Multiple linear regression analysis showed that the number of closed days, job title, stress level and whether being the only child or not were included in the regression equation. Conclusion The main influencing factors of fatigue in officers and soldiers during the COVID-19 epidemic include gender, job title, service age, educational background, stress level, number of closed days, and the impact of the epidemic on themselves. The number of closed days, job title, stress level and whether being the only child or not can effectively predict the fatigue status of officers and soldiers during closed management.

Objective To investigate the insomnia status and the related influencing factors of clinical nurses fighting against coronavirus disease 2019 (COVID-19) in Wuhan, so as to provide references for targeted psychological intervention during the prevention and control of the COVID-19 epidemic. Methods Convenience sampling method was conducted among clinical nurses fighting against COVID-19 in the Zhongnan Hospital of Wuhan University from Mar. 3 to 7, 2020. General information questionnaire, Athens insomnia scale (AIS) and patient health questionnaire (PHQ-9) were used, and multivariate linear regression analysis was conducted to analyze the influencing factors of insomnia in clinical nurses fighting against COVID-19. Results A total of 521 questionnaires were collected, including 504 valid ones, with an effective rate of 96.74%. The AIS score was 6 (3, 9), of which 352 (69.84%) had sleep disorder;the PHQ-9 score was 6 (2, 9), of which 292 (57.94%) had different degrees of depression. Professional title, total working time at epidemic frontline, whether the direct relatives diagnosed as COVID-19 or not, fear of the COVID-19 epidemic and depression level were included in the regression equation (all P＜0.05), which explained 56.2% of the total variation of insomnia in clinical nurses fighting against COVID-19. Conclusion Insomnia is common among clinical nurses fighting against COVID-19 in Wuhan, and the related influencing factors are complex. Nursing administrators should pay attention to the insomnia status of clinical nurses fighting against COVID-19, and all-round and effective interventions should be carried out in time, so as to improve the sleep quality of nurses.
