Background: The primary cause of treatment failure in patients with refractory or relapsed B-cell non-Hodgkin lymphoma (r/r B-NHL) is resistance to current therapies, and therapy-induced senescence (TIS) stands out as a crucial mechanism contributing to tumor drug resistance.Here, we analyzed SENEX/Rho GTPase Activating Protein 18 (ARHGAP18) expression and prognostic significance in doxorubicin-induced B-NHL-TIS model and r/r B-NHL patients, investigating its target in B-NHL cell senescence and the effect of combining specific inhibitors on apoptosis resistance in B-NHL-TIS cells.Methods: Raji cells were transfected with the human SENEX shRNA recombinant lentiviral vector (Sh-SENEX) and the empty vector negative (NC) to construct a stable transfection cell line with knockdown of SENEX.Effect of SENEX-silencing on B-NHL-TIS formation, cell function and cell cycle-related pathways was analyzed.Using doxorubicin (DOX)-inducible senescent B-NHL cells combined with the specific cyclin dependent kinase 4/6 (CDK4/6) inhibitor Palbociclib to observe that blocking CDK4/6 effects on TIS formation.SENEX expression of 21 B-NHL patients and 8 healthy controls were analyzed by qRT-PCR, and the correlation between its expression and clinical indicators were evaluated.Results: The downregulation of SENEX expression promotes G1-S phase transition and apoptosis while inhibiting cell proliferation, collectively suppressing the formation of TIS in B-NHL.Blockade of CDK4/6 promotes the DOX-induced G1 phase arrest to enhance TIS formation in B-NHL cells which can reverse the regulatory effect of silencing SENEX on B-NHL cell cycle regulation and senescence.The expression levels of SENEX were notably elevated in B-NHL patients compared to healthy controls, and Elevated expression levels of SENEX were associated with poor prognosis of B-NHL patients.Conclusions: SENEX enhances apoptosis resistance in B-NHL by inhibiting CDK4/6, thereby preventing G1-S phase transition and promoting TIS formation.

Many phytochemicals and their derived metabolites produced by plants are extensively employed in commercial goods, pharmaceutical products as well as in the environmental and medical fields.However, these secondary metabolites obtained from plants are in low amounts, and it is difficult to synthesize them at the industrial level.Despite these challenges, they may be utilized for a variety of medicinal products that are either available in the market or are being researched and tested.Secondary metabolites are complex compounds that exhibit chirality.Further, under controlled conditions with elicitors, desired secondary metabolites may be produced from plant cell cultures.This review emphasizes the various aspects of secondary metabolites including their types, synthesis, and applications as medicinal products.The article aims to promote the use of plant secondary metabolites in the management and treatment of various diseases.

A perspective review on the biosynthesis of plant-based secondary metabolites and their application as potent drugs

Background: Ferroptosis, a lipid peroxidation-mediated programmed cell death, is closely linked to tumor development, including prostate cancer (PCa).Despite established connections between ferroptosis and PCa, a comprehensive investigation is essential for understanding its impact on patient prognosis.Methods: A risk model incorporating four ferroptosis-related genes was developed and validated.Elevated risk scores correlated with an increased likelihood of biochemical recurrence (BCR), diminished immune infiltration, and adverse clinicopathological characteristics.To corroborate these results, we performed validation analyses utilizing datasets from both the Cancer Genome Atlas Cohort (TCGA) and the Gene Expression Synthesis Cohort (GEO).Moreover, we conducted further investigations into the pivotal gene identified in our model to explore its impact on tumor characteristics through cell proliferation and invasion assays, as well as animal studies conducted in vivo.Additionally, we conducted further experiments involving ferroptosis-related analysis to validate its association with ferroptosis.Results: The risk model demonstrated exceptional predictive capabilities for prognosis and therapeutic outcomes in PCa patients.Mitogen-activated protein kinase 9 (MAPK9) emerged as a crucial gene within the model.In vivo and in vitro experiments explored MAPK9's role in ferroptosis and its influence on tumor migration and proliferation. Conclusion:The findings provide a novel perspective for advancing ferroptosis exploration in PCa, bridging basic research and clinical applications.

MAPK9 as a therapeutic target: unveiling ferroptosis in localized prostate cancer progression

Mesenchymal stem cells (MSCs) have been proposed in regenerative medicine, especially for angiogenic purposes, due to their potential to self-renew, differentiate, and regulate the microenvironment.Peripheral vascular diseases, which are associated with reduced blood supply, have been treated but not cured.An effective therapy is to recover blood supply via vessel regeneration in the affected area, and MSCs appear to be promising for such conditions.Several studies aimed to explore the role of MSCs in performing angiogenesis and have revealed numerous potential methods to enhance MSC capacity in vessel formation.Efforts have been made to modify standard MSCs to optimize their prospective characteristics.Thus, understanding the mechanism underlying MSC angiogenic potential, and learning how to control their effect in the in vivo environment is critical for success in clinical settings.This review covers the angiogenesis process and discusses the role of some of the key angiogenic regulators.Possible techniques to enhance the angiogenic capacity of MSCs are also mentioned to suggest potential methods for therapeutic application.
