Radioactively labelled RNA transcripts made in vitro of various fragments from cDNA clones of poliovirus type 1 and of hepatitis A virus under the control of bacteriophage T7 or SP6 promoters have been evaluated for diagnostic purposes. The RNA transcripts were 2 orders of magnitude more sensitive as hybridization probes than corresponding cDNA preparations labelled by the nick translation procedure. A combination of hybridization analysis and sequence comparison showed that some regions of the genome of a number of enteroviruses are highly conserved, while others show very little homology; the general order of conservation is: 5'-non-coding greater than 3'-terminal greater than central (2C) greater than VP3 greater than VP1. The 350 bases of the poliovirus VP1 region were highly specific for that virus, while the 450-bases of the 5'NC region showed extensive cross-reaction with other enteroviruses. However, these probes did not hybridize with HAV, which was detected only by HAV-specific riboprobes. The transcripts have been successfully applied as hybridization probes in diagnostic tests on supernatants of infected cell culture lysates and in clinical samples, mainly in stool extracts.

Three peptides corresponding to residues 28-40, 138-154 and 380-387 of the coat protein of tomato bushy stunt virus (TBSV) were synthesized by the solid phase method and used to raise specific antibodies. These antibodies were used to follow the conformational changes that occur when TBSV particles swell under slightly alkaline conditions. Peptides 28-40 and 380-387 were found to correspond to continuous epitopes in the dissociated viral protein as well as in both compact and swollen virions. The region 138-154, which is also a continuous epitope of the monomeric protein, became accessible to antibody binding in the virion only when the particles were in the swollen state.

Accessibility of three continuous epitopes in tomato bushy stunt virus

An in situ ELISA was performed directly on the adherent cell monolayer in order to determine the susceptibility of herpes simplex virus isolates to acyclovir. Various fixation procedures and antisera conjugated to different enzymes were tested. The use of glutaraldehyde for fixation and betagalactosidase as a labelling enzyme was shown to give the best results. As with other currently used assays, 50% inhibitory doses were subject to an inoculum effect. The data obtained indicate that this assay is suitable for routine determination of herpes simplex virus susceptibility to antiviral drugs. Pour tester la sensibilité du virus de l'herpes simplex à l'acyclovir, nous avons réalisé un test ELISA directement sur monocouche cellulaire. Différentes méthodes de fixation et différents enzymes conjugués aux immunsérums ont été essayés. Les meilleurs résultats ont été obtenus avec le glutaraldéhyde comme fixateur et avec la beta-galactosidase comme enzyme. Comme pour les autres tests, les doses inhibitrices 50% varient en fonction de l'inoculum. Les résultats obtenus montrent que ce test est simple et rapide et qu'il est adapté à une détermination de routine de la sensibilité des virus herpes simplex aux antiviraux.

Evaluation of herpes simplex virus susceptibility to acyclovir using an enzyme-linked immunosorbent assay

Resident peritoneal macrophages obtained from If-1h and If-1l mice were induced in vitro with Newcastle disease virus. Then, 3, 5 and 7 h after induction, the cells were fixed and analysed for the presence of Mu IFN-beta mRNA by in situ hybridization, using a 35S-labelled Mu IFN-beta cDNA as a probe. The number of cells that were scored positive was the same in cultures derived from high responder If-1h and from low responder If-1l mice, and reached 100% of the cells present in the cultures. This result highly suggests that low responder mice have the same number of IFN-producing cells as high responders, and that the difference in production is not due to an increased number of producer cells in high responders.
