The Archives of Histology and Cytology provides prompt publication in English of original works on the histology and histochemistry of man and animals. The articles published are in principle restricted to studies on vertebrates, but investigations using invertebrates may be accepted when the intention and results present issues of common interest to vertebrate researchers. Pathological studies may also be accepted, if the observations and interpretations are deemed to contribute toward increasing knowledge of the normal features of the cells or tissues concerned. This journal will also publish reviews offering evaluations and critical interpretations of recent studies and theories.

A need for identifying ganglioside-positive cells with neuronal markers prompted us to establish a reliable method for double or triple immunostaining nervous tissues. Perfusion fixation with paraformaldehyde is typically performed for the routine immunostaining of various neuronal markers but is not suitable for immunostaining gangliosides. Acetone fixation of fresh cryosections is frequently used for ganglioside immunodetection; thus, we tested the effect of acetone treatment for unmasking the antigen epitope of gangliosides (acetone etching) on sections of paraformaldehyde-fixed nervous tissue from rats. Acetone etching significantly retrieved ganglioside immunoreactivity while preserving the immunoreactivity of neuronal markers. Various combinations of gangliosides and neuronal markers could be double-stained by the immunoenzyme method or triple-stained by the immunofluorescence method. This new method may provide additional information regarding the relationship between gangliosides and various neuronal markers from routinely paraformaldehyde-fixed nervous tissues, both freshly prepared specimens and those stocked in the laboratory.

Simultaneous immunohistochemical detection of gangliosides and neuronal markers in paraformaldehyde-fixed nervous tissues by acetone etching

The microstructure of lingual papillae on the dorsal surface of the tongue of adult Egyptian fruit bats was examined by light microscopy (LM) and scanning electron microscopy (SEM). This elongated tongue with a rounded apex is approximately 3 cm long -- including the 1.7cm length of the anterior free part of the tongue -- which facilitates considerable freedom of movement. The surface of the tongue has four types of lingual papillae: two types of mechanical papillae -- filiform and conical papillae, and two types of gustatory papillae -- fungiform and vallate papillae. Most numerous are filiform papillae with well developed keratinized processes represented by four morphological subtypes -- small, giant, elongated, and bifid papillae. Our observations showed the small and giant filiform papillae to be present in the anterior part of the tongue and tilted to the back of the tongue. In the posterior part of the tongue, the filiform papillae with elongated processes were arranged on each side of the tongue and oriented perpendicularly to the median line of tongue. This arrangement of filiform papillae is considered to be useful for the efficient uptake of semiliquid food as it can be collected toward the median line of the tongue. Gustatory fungiform papillae were distributed among filiform papillae on the border of the apex and the anterior part of the body of the tongue and also on the posterior part of the tongue, while three vallate papillae surrounded by conical papillae were found on the root of the tongue. There were also taste buds along the ducts of the posterior lingual glands in the posterior-lateral part of the tongue. These morphological features are discussed in relation to adaptation to food uptake in the Egyptian fruit bat.

The microstructure of lingual papillae in the Egyptian fruit bat (Rousettus aegyptiacus) as observed by light microscopy and scanning electron microscopy

High-speed atomic force microscopy (HS AFM) in ‘contact' mode was used to image at video rate the surfaces of both calcium hydroxyapatite samples, often used as artificial dental enamel in such experiments, and polished actual bovine dental enamel in both neutral and acidic aqueous environments. The image in each frame of the video of the sample was a few micrometers square, and the high-speed scan window was panned across the sample in real time to examine larger areas. Conventional AFM images of the same regions of the sample were also recorded before and after high-speed imaging. The ability of HS AFM to follow processes occurring in liquid on the timescale of a few seconds was employed to study the dissolution process of both hydroxyapatite and bovine enamel under acidic conditions. Buffered citric acid at pH values between 3.0 and 4.0 was observed to dissolve the surface layers of these samples. The movies recorded showed rapid dissolution of the bovine enamel in particular, which proceeded until the relatively small amount of acid available had been exhausted. A comparison was made with enamel samples that had been treated in fluoride solution (1 h in 300 ppm NaF, pH 7) prior to addition of the acid; the speed of dissolution for these samples was much less than that of the untreated samples. The HS AFM used an in-house designed and constructed high-speed flexure scan stage employing a push-pull piezo actuator arrangement. The HS AFM is able to follow the large changes in height (on the micrometer scale) that occur during the dissolution process.

High-speed atomic force microscopy of dental enamel dissolution in citric acid

Amyloid fibrils are primarily known in a pathogenic context for their association with a wide range of debilitating human diseases. Here we show a marine invertebrate (Entobdella soleae) utilizes functional amyloid fibrils comparable to those of a unicellular prokaryote (Escherichia coli). Thioflavin-T binding and Raman spectroscopy provided evidence for the presence of amyloid in the adhesive of Entobdella soleae. We elucidated that for these two very different organisms, amyloid fibrils provide adhesive and cohesive strength to their natural adhesives. Comparing the nanoscale mechanical responses of these fibrils with those of pathogenic amyloid by atomic force microscopy revealed that the molecular level origin of the cohesive strength was associated with the generic intermolecular β-sheet structure of amyloid fibrils. Functional adhesive residues were found only in the case of the functional amyloid. Atomic force microscopy provided a useful means to characterize the internal structural forces within individual amyloid fibrils and how these relate to the mechanical performance of both functional and pathogenic amyloid. The mechanistic link of amyloid-based cohesive and adhesive strength could be widespread amongst natural adhesives, irrespective of environment, providing a new strategy for biomimicry and a new source of materials for understanding the formation and stability of amyloid fibrils more generally.

Mechanically functional amyloid fibrils in the adhesive of a marine invertebrate as revealed by Raman spectroscopy and atomic force microscopy

The nerve growth factor-induced gene B-β (NGFI-Bβ, Nurr1) is a member of the nuclear receptor superfamily that is expressed predominantly in the central nervous system. We used an antibody against the human NGFI-Bβ to observe the protein expression in neuronal cells in the retina, cerebral neocortex, and midbrain of humans and rats. To provide further insight into the role of NGFI-Bβ in the differentiation of neuronal cells, we also examined the expression of NGFI-Bβ in rat ontogeny. A few cells in the midbrain showed the expression of NGFIBβ from 12 days of gestation, and NGFI-Bβ positive cells increased in the neocortex, claustrum, thalamus and hypothalamus in the subsequent fetal days. NGFI-Bβ-positive cells appeared in the inner nuclear layer of the retina at 18 days of gestation and also in the ganglion cell layer after birth. An immunohistochemical study on the expression of proliferating cell nuclear antigen (PCNA) demonstrated that NGFI-Bβ-positive cells were not proliferating cells. These findings suggest that NGFI-Bβ plays an important role during the postmitotic differentiation of neuronal cells in the brain and retina.

Summary. The aim of this study was to evaluate the distribution pattern of the different subtypes of the interstitial cells of Cajal (ICC), to understand their roles in gastrointestinal motility and interaction with the enteric nervous system depending on their location. Based on morphological analyses using the immunohistochemistry of whole mount preparations of mouse intestine, we investigated the normal local density and morphological features of ICC and statistically compared the number of the cells between the mesenteric and anti-mesenteric sides, and between the proximal and distal sites. Density gradients were shown in ICC associated with the deep muscular plexus, the myenteric plexus in the large intestine, and the longitudinal muscle. However, such changes in cell density were not always proportional to other subtypes, even in the same part of the intestine. As the pacemaker cells for intestinal motility, dense and seamless network connections were shown in ICC associated with the myenteric plexus in the small intestine and ICC associated with the submuscular plexus in the large intestine without any gradient of cell density, in contrast with the difference of morphological network patterns between proximal and distal sites. Thus, we conclude that the distribution patterns of ICC differ in cell density, morphological features, and network patterns, depending on the individual subtype and site of the intestine. Therefore, careful studies on the quantification and distribution of ICC under normal conditions should be important in diagnosing clinical disorders related to abnormal distribution of ICC.

Immunohistochemical characterization of interstitial cells of Cajal (ICC) in the mouse intestine using whole mount preparations – with particular reference to their density, morphological features, and network patterns

Summary. Neonatal hypoxic/ischemic (H/I) brain injury causes neurological impairment, including cognitive and motor dysfunction as well as seizures. Patterns of H/I injury-induced neuron death using rodent models are considered to be similar to the cases in human H/I encephalopathy. The participation of autophagy in neuron death has been a common feature in neonatal rodent models of H/I brain injury and human H/I encephalopathy when examined by immunochemical approaches for MAP1-LC3. This tendency has also been confirmed in neuronal tissue-specific Atg7 conditional knockout mice. However, while the current rat H/I model that is used for analyzing autophagy results in global damage to the ipsilateral hemisphere, it does not entirely reflect the neuropathological changes that appear in the neonatal mouse H/I model, in which the hippocampus is selectively damaged. The present study established a neonatal rat model of H/I injury with a milder ischemic insult, in which autophagy was involved in the hippocampal CA1 region after H/I injury when examined by electron microscopy, and by immunohistochemical and biochemical analyses of LC3.

Summary. Aquaporin-2 (AQP2) is a water channel protein that is trafficked between intracellular vesicles and the plasma membrane of kidney collecting duct cells upon vasopressin stimulation. Vasopressin changes the phosphorylation states of the AQP2 C-terminal serines (Sers), Ser256, Ser261, Ser264, and Ser269, in rats and mice, which is thought to play a role in controlling trafficking. Here, we focused on Ser269. We developed a specific antibody to Ser269-phosphorylated AQP2. Using immunofluorescence microscopy, we examined its localization in the rat kidney following injection of vasopressin and a vasopressin type 2 receptor-specific antagonist (OPC-31260). Ser269-phosphorylated AQP2 was almost undetectable in the water-loaded rat kidney, but was detected intracellularly soon after vasopressin injection, and then highly accumulated on the apical membrane of connecting tubule and collecting duct principal cells. In addition to the apical membrane, Ser269- phosphorylated AQP2 was also detected on the basolateral membrane of connecting tubule cells and inner medullary collecting duct principal cells. OPC-31260 injection following vasopressin stimulation caused internalization of AQP2, a pool of which was phosphorylated at Ser269. These results suggest that 1) AQP2 is phosphorylated at Ser269 intracellularly upon vasopressin stimulation and is rapidly trafficked to the plasma membrane, and 2) AQP2 can be internalized from the plasma membrane even if it remains phosphorylated at Ser269.

Summary. In Xenopus laevis Daudin the anlage of the carotid labyrinth, a baro- and chemoreceptor, appears in early premetamorphosis (NF stage 53) as a swelling of the third aortic arch at the point where the common carotid artery gives off the external carotid artery. A conspicuous accumulation of melanocytes preceeds the formation of the labyrinth maze which starts to grow at the end of metamorphic climax (NF stage 64) and continues so through juvenile age. In adult animals the carotid labyrinth displays a spherical to ovoid shape, has an average length of 1.4 mm, a width of about 0.8 mm and a height of about 0.4 mm. The maze vessels form by sprouting angiogenesis from the common carotid artery and the internal carotid artery. Scanning electron microscopy of vascular corrosion casts reveals that size, overall shape, surface morphology and vascular patterns of the carotid labyrinth vary greatly between individuals, whereby intussusceptive microvascular growth (= nonsprouting angiogenesis) and its facets model and contribute to the maturation of the labyrinth’s final vascular patterns. Although salivary glands are one of the target organs in graft-versus host disease (GVHD), mechanism of disease-related tissue injury has been poorly understood. To investigate it, we employed rat GVHD model including irradiation and donor cell transfer. Immunostaining revealed that submandibular and parotid, but not sublingual glands showed acinar reduction whereas ducts were almost intact. Ductal area of sublingual glands was significantly smaller than those of submandibular and parotid glands. Ductal epithelial cells upregulated expression of class II major histocompatibility complex antigens overtime. This expression preceded donor cell infiltration. Donor cell infiltration was preferentially found in ductal area at the early phase of the disease. While ductal epithelial cells exhibited upregulated mRNA expression of Th1-type chemokines, infiltrating donor cells exhibited their receptors CXCR3 and CCR5. These results suggest ductal epithelial cells have a pivotal role in the progression of GVHD.
