The precipitation evolution during ageing of a 2101 lean duplex stainless steel was investigated, revealing that the precipitate type and morphology depends on the nature of the grain boundary. Triangular M23C6 carbides precipitate only at γ/δ interfaces and rod-like Cr2N nitrides precipitate at both γ/δ and δ/δ interfaces. After 15min of ageing, the M23C6 size no longer evolves, whereas that of the Cr2N continues to evolve. For Cr2N, the morphology is maintained at γ/δ interfaces, whereas percolation occurs to form a continuous layer at δ/δ interfaces. By combining 2D and 3D characterisation at the nanoscale using transmission electron microscopy (TEM) and focused ion beam (FIB) tomography, a complete description of the precipitation evolution was obtained, including the composition, crystallographic structure, orientation relationship with the matrix phases, location, morphology, size and volume fraction.

M23C6 carbides and Cr2N nitrides in aged duplex stainless steel: A SEM, TEM and FIB tomography investigation.

The ultrastructural analysis of tobacco, potato and pepper tissues during infection with necrotic strains and the ordinary Potato virus Y strain of revealed the presence of virus inclusions not only in the epidermis and mesophyll but also in the vascular tissues. For the first time cytoplasmic inclusions were documented in companion cells and phloem parenchyma as well as in xylem tracheary elements. The ultrastructural features studied in this work consisted of mostly laminated inclusions (in the traverse and longitudinal section), which were frequently connected with enlarged cisternae of endoplasmic reticulum (ER) located in the direct vicinity of the cell wall attached to virus particles opposite to plasmodesmata. It was noticed that ER participates in synthesis and condensation of the PVY inclusions. During compatible interaction of tobacco and potato plants with PVY, amorphous and nuclear inclusions were observed. Such forms were not found in pepper tissues and potato revealing the hypersensitivity reaction to the infection with PVY necrotic strains. It was stated that the forms of cytoplasmic inclusions cannot serve as a cytological criterion to distinguish the potato virus Y strains and do not depend on host resistance level. Only in compatible interaction in Solanaceous plants tissues cytoplasmic inclusions were observed from the moment the morphological symptoms appeared. In the reaction of hypersensitivity, the inclusions were found on the 24th day following the infection with the PVY necrotic strains, whereas the symptoms were observed 3 days after the PVY infection.

Cytopathological potato virus Y structures during Solanaceous plants infection.

Wool fibre surfaces have been treated by solvent cleaning which leaves the native covalently bound surface lipid layer intact, and by alcoholic alkali which removes the lipid layer. The resultant surfaces have been analysed by atomic force microscopy (AFM), with particular emphasis on force-distance (F-d) methods. Methodologies were developed for investigation in situ in water of both the surface topography and the characteristics of the lipid layer. Longitudinal surface texturing was resolved in images of wool fibre surfaces in air; the texturing remained prominent after exposure to water. High resolution F-d curves revealed features associated with the lipid layer. A simple formalism was used to show that the layer had a thickness of a few nm, and an effective stiffness of some 0. 12+/-0.01N/m. Strong adhesive interactions, equivalent to a pressure of 0.1MPa, acted on the tip at the tip-to-substrate interface. The methodology and formalism are likely to be relevant in the broad field of thin-film analysis and for fibre technology.

Atomic force microscopy analysis of wool fibre surfaces in air and under water.

We present a theoretical analysis of the image formation in structured illumination wide-field fluorescence microscopy (SIWFFM). We show that the optically sectioned images obtained with this approach possess the optical sectioning strengths comparable to those obtained with the confocal microscope. We further show that the transfer function behaviour is directly comparable to that of the true confocal instrument. The theoretical considerations are compared with and confirmed by experimental results.

Electron-beam irradiation of α-Mn triggers dramatic microstructural transformations. Transmission electron microscopy (TEM) reveals localized thinning and MnO formation within the irradiated area. Reduced thermal conductivity due to thinning suggests significant local temperature rise by electron-beam irradiation. Finite element analysis (FEA) identifies Auger excitation as the dominant heating mechanism, surging temperatures to ∼2300 K with ultrafast cooling. Our findings indicate that the oxidation of Mn under electron-beam irradiation is primarily attributed to beam heating via Auger excitation, rather than defect formation through sputtering. This conclusion is supported by the fact that the maximum energy transferable from the incident electron beam in TEM is below the minimum displacement energy for Mn.

Electron-beam induced Mn oxidation in TEM: Insights into the heating effect of Auger excitation

HfO2 has been widely used in the electronics industry as a dielectric material with excellent properties. It has attracted much attention since HfO2 was first reported to be ferroelectric in 2011. With the continuous advancement of research, various methods such as oxygen vacancy control and interface control have been proven to be able to stabilize the metastable polar o-phase structure in doped hafnium oxide thin films. However, there are still some shortcomings in the relevant issues concerning non-doped hafnium oxide thin film materials. Here, polycrystalline non-doped HfO2 thin films were grown on SrTiO3 substrates by pulsed laser deposition (PLD). Atomic force microscopy investigation suggests that the surface roughness of HfO2 thin films increases as the film thickness increases. X-ray photoelectron spectroscopy analyses indicate that the HfO2 thin film has a high purity and contain only Hf4+ ions. The band gap of HfO2 was measured by valence EELS and UV-visible spectra. Atomic structures of the HfO2/SrTiO3 heterointerface have been studied by the aberration-corrected transmission electron microscopy and energy-dispersive X-ray spectroscopy. The HfO2/SrTiO3 heterointerface is atomically abrupt and incoherent. Our findings suggest that non-doped HfO2 films with o-phase structure through PLD technology.

On the effect of precession for magnetic differential phase contrast imaging

Organisms actively respond to shifts in their environment, and these responses are evident even among microinvertebrates like tardigrades. Encystment, regarded as a form of diapause, exemplifies a tardigrade's response to environmental change. Environmental cues and unidentified internal factors regulate this process in tardigrades. While it is known that some species can form cysts, our understanding of encystment in tardigrades remains limited, necessitating further research. We investigated selected morphological and physiological aspects of encystment to improve our understanding of the organisation and physiology of encysted animals. The data collected in this study were used to examine cellular organisation, overall morphology, and anatomy, including changes during cyst formation. We also explored the relationship between the body wall and somatic muscles. This paper provides a comprehensive overview of encysted animals' organisation, focusing on the integument and somatic muscles, as well as their role in shaping morphology during cyst formation. Additionally, the changes observed in storage cells and their significance in encystment are discussed. Despite time-dependent changes in the storage cells, our data do not support claims of organ histolysis as part of the typical changes occurring during encystment in the species we analysed.
