Critical Reviews in Eukaryotic Gene Expression

Tuberculosis (TB) remains a major threat to global health. Biomarkers derived from pathogen-host interaction can facilitate the monitoring of active TB. The recent progress regarding such biomarkers is summarized, including those can be used from serum, sputum, urine, or breath monitoring. A wide range of potential biomarkers such as protein antigens, cell-free nucleic acids, and lipoarabinomannose were compiled. The possible use of biomarkers for infection identification and monitoring drug efficacy are also presented.

Prognostic Values of Homocysteine and Potassium in Acute Ischemic Stroke Patients After Intravenous Thrombolysis With rt-PA

Lung adenocarcinoma (LUAD) is the leading cause of cancer-related deaths globally, with late diagnoses often resulting in poor prognoses. The extracellular matrix (ECM) plays a crucial role in cancer cell processes. Using big data from RNA-seq of LUAD, we aimed to screen ECM-related lncRNAs (long noncoding RNAs) to determine their prognostic significance. Our study analyzed the LUAD cohort from The Cancer Genome Atlas (TCGA). Univariate Cox analysis identified prognostic lncRNAs, and least absolute shrinkage and selection operator (LASSO) regression analysis, followed by multivariate Cox analysis, was used to construct a prognostic model. Kaplan-Meier and ROC curves evaluated the model's prognostic performance. A nomogram was created to predict 3-year survival. Enrichment analysis identified biological processes and pathways involved in the signature. Correlations with the tumor microenvironment (TME) and tumor mutation burden (TMB) were analyzed, and potential drug sensitivities for LUAD were predicted. We initially identified 218 ECM-associated genes and 427 ECM-associated lncRNAs within the TCGA LUAD cohort. Subsequent univariate Cox regression analysis selected 26 lncRNAs with significant prognostic value, and an overall survival (OS)-based LASSO Cox regression model further narrowed this to 14 lncRNAs. Multiple Cox regression analyses then distilled these down to 8 critical lncRNAs forming our prognostic risk signature. Nomograms accurately predicted survival. Finally, several potential therapeutic drugs, including afatinib and crizotinib, were identified. Big data analysis established a prognostic signature that predicts survival and immunization in LUAD patients, providing new insights into survival and treatment options.

Identification of a Risk Signature and Immune Cell Infiltration Based on Extracellular Matrix-Related lncRNAs in Lung Adenocarcinoma

The purpose of this study is to identify the key regulatory genes related to the inflammatory response of esophageal adenocarcinoma (EAC) and to find new diagnosis and therapeutic options. We downloaded the dataset GSE72874 from the Gene Expression Omnibus database for this study. Weighted gene co-expression network analysis (WGCNA) and differentially expressed genes (DEGs) analysis were used to find common inflammatory response-related genes (IRRGs) in EAC. The relationship between normal and tumor immune infiltration was analyzed using an online database of CIBERSORTx. Finally, 920 DEGs were identified, of which 5 genes were key IRRGs associated with EAC, including three down-regulated genes GNA15, MXD1, and NOD2, and two down-regulated genes PLAUR and TIMP1. Further research found that GNA15, MXD1, and NOD2 were down-regulated, PLAUR and TIMP1 were up-regulated in Barrett's esophagus (BE). In addition, we found that the expression of GNA15 and MXD1 in normal esophageal squamous epithelial cells decreased after ethanol treatment, while the expression of PLAUR and TIMP1 increased after ethanol treatment. Compared with normal esophageal tissue, immune cells infiltrated such as plasma cells, macrophages M0, macrophages M1, macrophages M2, dendritic cells activated, and mast cells activated were significantly increased in EAC, while immune cells infiltrated such as T cells CD4 memory resting, T cells follicular helper, NK cells resting, and dendritic cells resting were significantly reduced. The receiver operating characteristic curve indicated that GNA15, MXD1, NOD2, PLAUR and TIMP1 expression had a performed well in diagnosing EAC from healthy control. GNA15, MXD1, NOD2, PLAUR and TIMP1 were identified and validated as novel potential biomarkers for early diagnosis and may be new molecular targets for treatment of EAC.

Identification of a Diagnosis and Therapeutic Inflammatory Response-Related Gene Signature Associated with Esophageal Adenocarcinoma

Extensive research has recently been conducted to investigate the regulating impact of exosomal circular RNAs (circRNAs) in throughout the development of multiple malignancies. Nevertheless, there is still much to learn about the biological roles and underlying mechanisms of exosomal circRNAs in colorectal cancer (CRC). Exosomes (exo) were isolated from blood samples and CRC cells by differential centrifugation. In addition, the competitive endogenous RNA (ceRNA) mechanism of circ_001860 in CRC was determined through Starbase and dual-luciferase reporter gene experiments. Gain and loss of function experiments verified the regulatory effect of circ_001860/miR-582-5p/ZEB1 on the malignant phenotype of CRC cells. The therapeutic effect of circ_001860 on CRC xenograft tumor model was explored through mouse experiment. circ_001860 was significantly enriched in exo isolated from CRC blood samples and CRC cells. circ_001860 can be transported into CRC cells via exo. Through competitive binding to miR-582-5p, circ_001860 increased ZEB1, thereby facilitating tumor formation in vivo as well as stimulating CRC cell proliferation and metastasis in vitro. Through the miR-582-5p/ZEB1 axis, exosomal circ_001860 enhanced the advancement of CRC. This finding may offer non-invasive biomarkers for clinical screening and diagnosis of CRC patients.
