The topics in plant science encompass fundamental research in taxonomy, earth botany, genetics, and experimental botany, covering areas such as cellular, whole plant, and community physiology. Abstract The aim of this study was to identify the role of field margin habitats in preserving the diversity and abundance of ground beetle assemblages, including potentially entomophagous species and those with conservation status in Poland . Research material was collected in 2006–2007 in four types of margin habitats – a forest, bushes, ditches and in two arable fields. Insects were captured into pitfalls, without preservation liquid or bait added to the traps . Traps were inspected twice a week, between May and August, and one sample was a weekly capture . In field margin habitats the most abundant species were Limodromus assimilis , Anchomenus dorsalis , Pterostichus melanarius and Carabus auratus . A lower abundance of species was noted on fields, with dominant Poecilus cupreus and P. melanarius . The group of zoophagous carabids found in our study includes 30 species from field margin habitats, i.e. 37.5% of all captured Carabidae taxa and 58.3% of all specimens. The share of aphidophagous species was 84.9% among bushes, 86.7% near ditches, and 88.0% in the forest habitat. Several species captured during the study are under protection in Poland. These include the partly protected Carabus convexus , which also has the status of near threatened species, the partly protected Calosoma auropunctatum , and Broscus cephalotes . Considering all the investigated field margin habitats, ground beetles were most numerous in the oak-hornbeam habitat, defined as bushes, formed predominantly by Prunus spinosa , Crataegus leavigata , Sambucus nigra and Rosa canina . Thus, this habitat was the most important reservoir/refugium for the ground beetles.

Ground beetles (Carabidae) of field margin habitats

Following the sequencing of rice genome, the functional analysis of unidentified genes is gaining wide importance. Mutant isolation is one of the effective ways to isolate and clone the target genes and analyze their functions. To find the various mutants in the same genetic background, seeds of Oryza sativa cv. Nipponbare were treated with ethyl methane sulphonate (EMS). A total of 1056 mutants were screened for five categories in M2 generation with the seedling frequency of 26.29‰ at three-leaf stage, but only 264 mutants were verified in M3 generation with a frequency of 6.57‰. Among the mutants verified in M3 generation, the frequency of leaf mutation was the highest (2.22‰), followed by seedling height (1.74‰) and the abiotic stress tolerance mutant (1.47‰). Nineteen characteristic mutations, including a big group of abiotic stress tolerant mutants such as herbicide resistant, salt tolerant and drought tolerant were identified at this stage. By observation of rice growth characteristics at different developmental stages, another 220 mutants have been isolated and verified in the M3 generation with the mutant frequency of 53.9‰ covering about 28 mutant traits. Among those identified, the highest frequencies were obtained for appearance of brown rice mutant with 18.37‰, followed by panicle mutant with 13.47‰, and grain mutants with 9.06‰. All the mutants screened above were suitable for gene function analysis and for utilization in agronomy.

The screening of mutants and construction of mutant library for Oryza sativa cv. Nipponbare via ethyl methane sulphonate inducing

Two populations of cogongrass [Imperata cylindrica (L.) Raeuschel], one from the saline regions of the Salt Range and the other from the non-saline regions of Faisalabad were assessed for salinity tolerance on the basis of some key morphological and physiological attributes. It was hypothesized that the tolerant population from the Salt Range must have developed some specific structural modifications, which are responsible for its better survival under high salinities. These adaptive components can be effectively used in modern technologies for improving degree of tolerance of other sensitive crops. The population from the Salt Range markedly excelled the Faisalabad population in terms of growth and physiological attributes measured in this study. The Faisalabad population of I. cylindrica was unable to survive at the highest salt level (200 mM NaCl). The tolerance of the Salt Range population to salt stress was found to be related to high accumulation of organic osmotica, particularly total free amino acids and proline as well as Ca2+ in the shoot. The distinctive structural modifications in the Salt Range population were found to be enhanced succulence, well-developed bulliform cells in leaves and smaller stomatal area.

The commercially available genetically modified plants authorized worldwide and therefore the target sequences for molecular detection of genetically modified organisms (GMOs) are ever-increasing. The European Union has implemented a set of very strict procedures for approval to grow, import and/or utilize GMOs as food or food ingredients. As a result, GMO laboratories and food production industry currently are forced to apply different methods to test raw material and complex processed food products. Three exogenous genes (the 35 s promoter of the cauliflower mosaic virus (35 s), nos terminator from Agrobacterium tumefaciens (nos), and the neomycin phosphotransferase II (nptII) gene) are commonly used in GMO detection. In this paper, a multiplex quantitative real-time PCR (qPCR) system was developed which allows simultaneously detection of the three exogenous genes in one reaction tube. The determined limits for the multiplex qPCR assays were 4 copies/reaction in maize samples. The specificity of the assays was demonstrated to be 100% according to the detection results of 23 genetically modified (GM) crops and 97 complex processed food products. The validation data show the individual PCR efficiency was accredited with negligible impacts between three detection channels in 7500 fluorescence quantitative PCR machine. These results indicate that this high-throughput multiplex qPCR method which combined with a reference gene is feasible for screening of GMOs, even for the processed food.
