Photochemistry of 4-phenyl-1,2,3-thiadiazole (PT) and 4,5-diphenyl-1,2,3-thiadiazole (DPT) in solution was studied at room temperature using UV-vis and IR transient absorption spectroscopies ( λ _ex = 266 nm). Ultrafast techniques show a very fast rise (<0.3 ps) of thiirene and thioketene species, formed from 1,2,3-thiadiazoles in the singlet excited state. The remarkable unimolecular stability of thiirenes in solution is observed. On a millisecond time scale thiirenes with phenyl substituents undergo an intermolecular reaction (dimerization of thiirene–thioketene complexes) leading to 1,3-dithiole derivatives.

Photochemical formation of thiirene and thioketene in 1,2,3-thiadiazoles with phenyl substituents studied by time-resolved spectroscopy

The comparative photostability of curcumin 1, and two non-phenolic curcuminoids: 1,7-diphenyl-1,6-heptadiene-3,5-dione 2 (unsubstituted curcumin) and dimethylcurcumin 3 in non-degassed dilute solutions (approximately 3-5 x 10(-5) mol l(-1)) has been established by UV-visible absorption spectroscopy; disappearance quantum yields were measured. The similar behavior of the three studied curcuminoids is indicative of only a moderate role of phenol groups in the photodegradation process. Structural analysis of the photodegradation products of compound 2 in more concentrated solution (approximately 3.6 x 10(-3) mol l(-1)) shows formation of benzaldehyde, cinnamaldehyde, 2'-hydroxy-5',6'-benzochalcone 4, flavanone 5 and some other unidentified photoproducts. Flavanone 5 is formed by irradiation of chalcone 4. It represents a unique example of photochemical conversion of a diarylheptanoid molecule into a flavonoid, another very important class of natural products.

Transcranial photobiomodulation for neurodevelopmental disorders: a narrative review

Photobiomodulation (PBM) is a treatment method based on research findings showing that irradiation with certain wavelengths of red or near-infrared light has been shown to produce a range of physiological effects in cells, tissues, animals and humans. Scientific research into PBM was initially started in the late 1960s by utilizing the newly invented (1960) lasers, and the therapy rapidly became known as “low-level laser therapy”. It was mainly used for wound healing and reduction of pain and inflammation. Despite other light sources being available during the first 40 years of PBM research, lasers remained by far the most commonly employed device, and in fact, some authors insisted that lasers were essential to the therapeutic benefit. Collimated, coherent, highly monochromatic beams with the possibility of high power densities were considered preferable. However in recent years, non-coherent light sources such as light-emitting diodes (LEDs) and broad-band lamps have become common. Advantages of LEDs include no laser safety considerations, ease of home use, ability to irradiate a large area of tissue at once, possibility of wearable devices, and much lower cost per mW. LED photobiomodulation is here to stay.

Photodynamic Therapy (PDT) offers a minimally invasive approach for treating various health conditions, employing a photosensitizer (PS) and specific light. Recent enhancements make PDT outpatient-friendly and less discomforting. Effectiveness hinges on selecting the appropriate PS. This article delves into natural and synthetic PSs, emphasizing the rising interest in natural alternatives for their safety. It explores their mechanisms, characteristics, and applications, offering insights into their potential contributions to advancing PDT. This extensive review delves into the preclinical and clinical landscape of natural PSs for PDT, shedding light on their diverse applications and promising outcomes. Compounds like curcumin, piperine, riboflavin, psoralen, hypericin, and others show significant potential in preclinical in vitro studies across various cell lines. In vivo, these photosensitizers prove effective against skin tumors, carcinomas, and sarcomas, inducing apoptosis, autophagy, and ROS generation for therapeutic efficacy. The review underscores the critical role of proper dosing and monitoring in balancing therapeutic benefits and risks. It highlights the advantages and limitations of natural PSs, emphasizing their specific targeting, bioavailability, and limited side effects. The future of PDT holds promising breakthroughs, taking from some evidence like Bergamot oil in nanostructured lipid carriers for dermatological conditions. Second-generation photosensitizer Tookad shows potential in prostate cancer treatment, while Tripterygium wilfordii Hook. F. emerges as an antimicrobial PDT source etc. Thus, environmental concerns in PDT prompt a shift to plant extracts for PS purification. The evidence-supported focus on natural PSs establishes this article as a key resource for advancing natural compounds in PDT and their therapeutic applications.

Abstract Higher prevalence of multiple sclerosis at higher latitudes is associated with reduced sunlight during childhood. Alterations in inflammatory Th17 and regulatory T cells (Treg) are associated with autoimmunity. In Hobart, Australia (latitude 42.8° south), thirteen girls (aged 12–13) and sixteen women had blood collected in the evening in daylight in February/March, (post 15 h daylight) and at the same time in the dark in August/September (post 9 h daylight). Height and weight were measured. Participants completed online surveys prior around menstruation, sleep, exercise and time outside. Proportions of Th17 (CD4 + , CXCR3–, CCR4 + , CCR6 + , CD161 +), total Treg (CD4 + , CD25 + , CD127low), naïve Treg (CD45RA + , CD4 + , CD25 + , CD127low) and memory Treg (CD45RA low, CD4 + , CD25 + , CD127low) were enumerated by flow cytometry (Cytek Aurora). Hours spent outside was greater in summer than winter (12.5 v 10.5, p = 0.0003). The %Treg/CD4 + was higher in summer than winter (7.3% vs 7%, p = 0.004) including memory Treg (3.1% vs 2.9%, p = 0.02) for all females ( n = 29), and naïve Treg were 13% higher in summer in women (3.2% vs 2.8%, p = 0.0009), whereas %Th17 remained unchanged. In women, a negative correlation between the number of hours spent outside in summer and %Th17 was observed ( r = – 0.53, p = 0.035). The %Th17 cells were higher in women than girls (4.9% vs 3.1%, p = 0.001), whereas girls had a higher total %Treg (7.6% vs 6.7%, p = 0.005), consisting of a higher naïve Treg (5.5% vs 3.7%, p = 0.0001) while the women had a higher %memory Treg (3.4% vs 2.6%, p = 0.0001). These light-dependent seasonal differences may influence immune development in adolescents. Graphical abstract

This review aims to provide the readership with an overview of the applications of the time-resolved spectroscopic techniques (often combined with steady-state measurements) in the elucidation of the mechanism of the photochemical behaviour of diaryl- and triarylamines. Such derivatives undergo cyclization to the carbazoles or ionization to the corresponding amine radical cations depending on both the nature of the starting substrates and the reaction conditions. In this context, time-resolved spectroscopy techniques allowed for the detection and the full characterization of all the excited states and the transients involved in these processes. The main characteristics of such species, including lifetime, time-resolved absorption spectra, quantum yield values and rate constants of formation and decay in both homogeneous and micro-heterogeneous environment (e.g. aqueous surfactant solutions) are thus summarized in the presented paper.

On the application of nanosecond laser flash photolysis to the investigation of the photoreactivity of di and triarylamines

The study aimed to assess the impact of combining potassium iodide (KI) with methylene blue (MB) in antimicrobial photodynamic therapy (aPDT) within an oral biofilm formed in situ. A single-phase, 14 days in situ study involved 21 volunteers, who wore a palatal appliance with 8 bovine dentin slabs. These slabs were exposed to a 20% sucrose solution 8 times a day, simulating a high cariogenic challenge. Following the intraoral phase, the biofilms formed on the slabs were randomly assigned to the treatments: C (0.9% NaCl); CHX (0.2% chlorhexidine); KI (75 mM KI); MBKI (0.005% MB + 75 mM KI); L (0.9% NaCl + red laser 660 nm, 18 J, 180 s); LMB (0.005% MB + laser); LKI (75 mM KI + laser); LMBKI (0.005% MB + 75 mM KI + laser). The treated biofilms were collected, diluted, and incubated to assess cell viability (CFU/mL) for total microorganisms, total lactobacilli, total streptococci, and mutans streptococci. Data were subjected to analysis using the Friedman test, followed by the Dunn test (α = 0.05). LMBKI group exhibited a noteworthy decrease in the viability of all microorganisms in comparison to groups C, KI, MBKI, MB, L, LMB, and LKI (p < 0.0001), and demonstrated a comparable reduction to the CHX group (p > 0.99). The combination of KI with MB in aPDT may be advocated as a non-invasive technique for diminishing the viability of polymicrobial oral biofilms, thereby aiding in the management of dental diseases.
