Abstract Euplotes woodruffi Gaw 1939 is a striking ciliate species commonly used in many fields of protistology. Euplotes parawoodruffi Song and Bradbury 1997 is separated from E . woodruffi by several morphological differences (e.g. the absence of pre‐oral pouch) and in particular the brackish habitat (vs. freshwater). However, recent molecular data dispute the separation of E . woodruffi into two species. Based on a brackish isolate from the southeast of China, we investigated the morphology, molecular phylogeny, and adaptation of E . woodruffi using living observation, protargol impregnation, gene sequencing, and cultivation experiments. The morphological variability present in our isolate encompasses the previous descriptions of E. woodruffi and E. parawoodruffi . The pre‐oral pouch, which occurs in living cells of freshwater isolates, is also recognizable in the brackish specimens. Laboratory experiments suggested that the brackish isolate could acclimatize and reproduce in both brackish and fresh water. The molecular analyses using 13 available small subunit ( SSU ) rRNA gene sequences showed that the minimum pairwise sequence similarity is as high as 99.2% and nine sequences from both brackish and freshwater isolates are even 100% matched. Therefore, we conclude that E . parawoodruffi is a junior synonym of E . woodruffi .

Periphytic ciliates play a vital role in the material cycle and energy flow of microbial food web, however, their taxonomy and biodiversity are inadequately studied given their high species richness. Two new and one little known species, viz. Derouxella lembodes gen. et sp. nov., Cyrtophoron multivacuolatum sp. nov., and Cyrtophoron apsheronica Aliev, 1991, collected from coastal waters of China, were investigated using modern methods. Derouxella gen. nov. can be recognized by having dorsoventrally flattened body, a podite, one fragmented preoral kinety, two parallel circumoral kineties, and somatic kineties progressively shortened from right to left. Morphological classification and phylogenetic analyses based on nuclear small subunit ribosomal RNA (nSSU rRNA) and mitochondrial small subunit ribosomal RNA (mtSSU rRNA) gene sequence data inferred that Derouxella gen. nov. occupies an intermediate position between Hartmannulidae and Dysteriidae. Cyrtophoron multivacuolatum sp. nov. is characterized by large body size, the numbers of somatic kineties and nematodesmal rods, and having numerous contractile vacuoles. The genus Cyrtophoron and the poorly known species C. apsheronica were redefined. Even with the addition of newly obtained nSSU rRNA and mtSSU rRNA gene sequences of Cyrtophoron, the family Chlamydodontidae was still recovered as a monophyletic group, the monophyly of Cyrtophoron was supported too.

New contributions to the Cyrtophoria ciliates (Protista, Ciliophora): Establishment of new taxa and phylogenetic analyses using two ribosomal genes

ABSTRACT. Kudoa thyrsites (Myxozoa: Multivalvulida) is a cosmopolitan marine parasite of fishes associated with post‐mortem tissue degradation. Financial losses incurred as a result of these infections are of concern to commercial fisheries. There is conflicting evidence whether K. thyrsites represents a cryptic species complex. Myxospore morphology is very similar for K. thyrsites across its range, but preliminary genetic analyses show some differences. Kudoa thyrsites and the morphologically similar Kudoa histolytica were examined from hosts in British Columbia, Canada, Oregon, USA, Chile, England, South Africa, Australia, and Japan. We compared myxospore morphology and DNA sequences of heat shock protein 70 and the small subunit, large subunit, and internal transcribed spacer 1 of the ribosomal DNA. There was some morphological variation between regional representatives, inconsistent with genetic analyses. Phylogenetically, major separations correlated to four broad geographic regions: Japan, Australia, eastern Pacific, and eastern Atlantic. Within these regions there was little additional genetic structure. These data are evidence for regional subdivision of K. thyrsites suggesting global transplantation of fishes has yet to homogenize these distinctions. Within regions, parasite gene flow appears to be high between host species, suggesting little host specificity and minimal cryptic speciation. Our data also indicate that K. histolytica is not a valid species, as it was morphologically and genetically indistinguishable from K. thyrsites.

Phylogeography of the Cosmopolitan Marine Parasite Kudoa thyrsites (Myxozoa: Myxosporea)

Abstract Fish species around the world are parasitized by myxozoans of the genus Kudoa, several of which infect and cause damage of commercial importance. In particular, Kudoa thyrsites and Kudoa amamiensis infect certain cultured fish species causing damage to muscle tissue, making the fish unmarketable. Kudoa thyrsites has a broad host and geographic range infecting over 35 different fish species worldwide, while K. amamiensis has only been reported from a few species in Japanese waters. Through morphological and molecular analyses we have confirmed the presence of both of these parasites in eastern Australian waters. In addition, a novel Kudoa species was identified, having stellate spores, with one polar capsule larger than the other three. The SSU rDNA sequence of this parasite was 1.5% different from K. thyrsites and is an outlier from K. thyrsites representatives in a phylogenetic analysis. Furthermore, the spores of this parasite are distinctly smaller than those of K. thyrsites, and thus it is described as Kudoa minithyrsites n. sp. Although the potential effects of K. minithyrsites n. sp. on its fish hosts are unknown, both K. thyrsites and K. amamiensis are associated with flesh quality problems in some cultured species and may be potential threats to an expanding aquaculture industry in Australia.

First Report of Three Kudoa Species from Eastern Australia: Kudoa thyrsites from Mahi mahi (Coryphaena hippurus), Kudoa amamiensis and Kudoa minithyrsites n. sp. from Sweeper (Pempheris ypsilychnus)

Abstract As chloroplast‐stealing or “kleptoplastidic” lineages become more reliant on stolen machinery, they also tend to become more specialized on the prey from which they acquire this machinery. For example, the ciliate Mesodinium rubrum obtains &gt; 95% of its carbon from photosynthesis, and specializes on plastids from the Teleaulax clade of cryptophytes. However, M. rubrum is sometimes observed in nature containing plastids from other cryptophyte species. Here, we report on substantial ingestion of the blue‐green cryptophyte Hemiselmis pacifica by M. rubrum , leading to organelle retention and transient increases in M. rubrum 's growth rate. However, microscopy data suggest that H. pacifica organelles do not experience the same rearrangement and integration as Teleaulax amphioxeia 's. We measured M. rubrum 's functional response, quantified the magnitude and duration of growth benefits, and estimated kleptoplastid photosynthetic rates. Our results suggest that a lack of discrimination between H. pacifica and the preferred prey T. amphioxeia (perhaps due to similarities in cryptophyte size and swimming behavior) may result in H. pacifica ingestion Thus, while blue‐green cryptophytes may represent a negligible prey source in natural environments, they may help M. rubrum survive when Teleaulax are unavailable. Furthermore, these results represent a useful tool for manipulating M. rubrum 's cell biology and photophysiology.

Abstract Thecate amoebae play important roles in terrestrial and aquatic ecosystems. This study introduces a novel thecofilosean amoeba from Arctic and Antarctic sea sediments. Phylogenetic analysis based on the 18S rDNA sequence places it in the family Chlamydophryidae (order Tectofilosida, class Thecofilosea). However, the novel organism exhibits a significant genetic divergence and distinct morphology from its closest relatives, prompting us to erect the novel genus Katarium with its type species Katarium polorum . K. polorum is a consumer of diatoms and prokaryotes, indicating an important role in nutrient cycling in the polar marine food webs.

Human babesiosis is a malaria-like, tick-borne infectious disease with a global distribution. Babesiosis is caused by intraerythrocytic, apicomplexan parasites of the genus Babesia. In the United States, human babesiosis is caused by Babesia microti and Babesia duncani. Current treatment for babesiosis includes either the combination of atovaquone and azithromycin or the combination of clindamycin and quinine. However, the side effects of these agents and the resistance posed by these parasites call for alternative approaches for treating human babesiosis. Proteases play several roles in the context of parasitic lifestyle and regulate basic biological processes including cell death, cell progression, and cell migration. Using the SYBR Green-1 assay, we screened a protease inhibitor library that consisted of 160 compounds against B. duncani in vitro and identified 13 preliminary hits. Dose response assays of hit compounds against B. duncani and B. microti under in vitro conditions identified five effective inhibitors against parasite growth. Effect of protease inhibitors on the intraerythrocytic development of Babesia microti and Babesia duncani, the causative agents of human babesiosis

Amyloodinium ocellatum is a protozoan parasite that causes amyloodiniosis in marine and brackish water fish, threatening global aquaculture. The present study investigates the morphology and ultrastructure of the free-living stages of A. ocellatum (tomont and dinospore) using light microscopy (LM), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Dinospores measured 13.03-19.66 μm in length, 12.32-18.71 μm in width, and were laterally flattened. Dinospores had a transverse flagellum for propulsion and a longitudinal flagellum for direction control. The cyst wall had three distinct layers and included cellulose. The outer wall was coated with numerous bacteria. The orange-red speckled eyespot was observed all tomont developmental stages and in the dinospore of A. ocellatum. Tomonts proliferation required successive nuclear division, the formation of new cyst walls, and cytoplasmic segregation. The cytoplasm comprises mainly the matrix, organelles, and inclusions. The matrix was grainy and evenly distributed. In addition to organelles, including mitochondria with tubular cristae, Golgi apparatus, and endoplasmic reticulum, the cytoplasm had starch grains and lipid droplets as inclusions. The A. ocellatum cells lacked chloroplasts. This study provides the first ultrastructural view of the cytoplasmic structure of the free-living stages of A. ocellatum.

Abstract Protists show diverse lifestyles and fulfill important ecological roles as primary producers, predators, symbionts, and parasites. The degradation of dead microbial biomass, instead, is mainly attributed to bacteria and fungi, while necrophagy by protists remains poorly recognized. Here, we assessed the food range specificity and feeding behavior of the algivorous flagellate Orciraptor agilis (Viridiraptoridae, Cercozoa) with a large‐scale feeding experiment. We demonstrate that this species is a broad‐range necrophage, which feeds on a variety of eukaryotic and prokaryotic algae, but fails to grow on the tested fungi. Furthermore, our microscopic observations reveal an unexpected flexibility of O. agilis in handling food items of different structures and biochemistry, demonstrating that sophisticated feeding strategies in protists do not necessarily indicate narrow food ranges.
