Background Depression is a widespread and complex mental disorder that is challenging to treat. The unpredictable chronic mild stress (UCMS) model, based on stress diathesis, is used to study depression because antidepressants often reverse its effects, providing predictive validity. However, the mechanisms behind UCMS are not fully understood. AMP-activated protein kinase (AMPK) is essential for regulating neuronal energy metabolism, and disruptions in its activation can impair brain function and synaptic integrity. Teneligliptin, a dipeptidyl peptidase-4 (DPP-4) inhibitor with anti-inflammatory, antioxidant, autophagy-modulating, and antiapoptotic properties, has not been studied for its potential to activate AMPK by inhibiting DPP-4, stimulating glucagon-like peptide-1, and decreasing the mammalian target of rapamycin pathway, which may enhance neuroprotection and neurogenesis. This study is the first to show teneligliptin’s potential therapeutic effect on UCMS-induced depression in mice by targeting the AMPK signaling pathway. Objective The current study was performed to assess the behavioral, oxidative stress biomarkers, neurotransmitter assay, and histopathology of the brain in UCMS-induced depression in mice. Materials and methods Mice were divided into five groups: group I (normal control, received saline), group II (UCMS model with 1 week of stress), group III (UCMS+fluoxetine 10 mg/kg daily for 4 weeks), and groups IV and V (UCMS+teneligliptin 30 mg/kg and 60 mg/kg, respectively, for 4 weeks). Body weight, forced swim test, tail suspension, locomotor activity, oxidative biomarkers (malondialdehyde, reduced glutathione, superoxide dismutase, nitrite levels), neurotransmitters (dopamine, serotonin), and brain histopathology were assessed to evaluate antidepressant activity. Results and conclusion Mice treated with teneligliptin (30 and 60 mg/kg) in the UCMS model showed significant weight loss compared with the disease group. They also exhibited reduced immobility in forced swim and tail suspension tests and increased locomotor activity. Significant changes were noted in oxidative stress biomarkers and neurotransmitters (dopamine, serotonin), with improved histopathological conditions. In conclusion, teneligliptin improved behavioral and antidepressant effects by reducing oxidative stress and increasing dopamine and serotonin levels. As a DPP-4 inhibitor, it may activate AMPK in the brain, lower DPP-4 enzyme levels, and enhance glucagon-like peptide-1-R and mammalian target of rapamycin, potentially promoting neurogenesis and neuroprotection, thus being beneficial for depression treatment.

Potential therapeutic effect of teneligliptin on unpredictable chronic mild stress-induced depression in mice: targeting the role of AMP-activated protein kinase signaling pathway

Background Saccharomyces cerevisiae strains were isolated from various sources and studied due to their significant potential as probiotics for treating different diseases. They act as adjuvants against gastrointestinal tract disorders, including bowel disease and various types of diarrhea in humans and animals. Consequently, identifying different strains of S. cerevisiae and confirming their safe consumption by humans and animals became crucial. Objective This study aims to isolate, identify, and characterize 36 S. cerevisiae isolates from different animal feed sources. The primary goal is to examine the diversity among these isolates. Materials and methods Yeast isolation from feed involved using Rose Bengal agar, which was incubated at a temperature of 30°C for 48 h. Isolate identification was performed using an internal transcribed spacer and S. cerevisiae -specific primers. Molecular markers, specifically sequence-related amplified polymorphism (SRAP) and inter-simple sequence repeat (ISSR), were used to determine the genetic diversity. Polymorphic information content was estimated, and genotype-specific markers were detected. A phylogenetic relationship was established based on data obtained from the molecular markers. Results and conclusion S. cerevisiae- specific primers confirmed that all 36 yeast isolates belonged to the S. cerevisiae species. Molecular identification was achieved through amplification of internal transcribed spacer sequences, followed by sequencing and database matching. Phylogenetic analysis further confirmed their classification as S. cerevisiae . Genetic diversity among the 36 strains was assessed using SRAP and ISSR markers, which show 23.4 and 30 polymorphism percentages, respectively. The results revealed that 26 SRAP and 18 ISSR genotype-specific markers were identified for the 36 S. cerevisiae isolates. The cluster analyses indicated that the strains were separated into 20 distinct clusters based on their genetic similarities. These findings demonstrate the effectiveness of S. cerevisiae -specific primers, molecular identification techniques, and genetic marker analyses in characterizing and understanding the genetic diversity and relatedness within a population of S. cerevisiae isolates.

Background Syndromic obesity is characterized by a specific set of associated clinical features, but the similarity between different obesity syndromes can make it hard to diagnose accurately. Prader-Willi syndrome (PWS) is the most common type of syndromic obesity. A variety of molecular and cytogenetic techniques may be needed for the diagnosis of obesity. Multiplex ligation-dependent probe amplification (MLPA) helps by allowing the study of multiple specific genetic regions at once. This makes it an effective screening tool for large groups of patients who may have deletions or duplications in specific genes. Objective We aim to establish a precise diagnostic scheme for early diagnosis that yields proper early intervention to prevent the development of morbid obesity and intellectual disability, which render a great burden on both health services and the families of the patients. Materials and methods Combined cytogenetic fluorescence in-situ hybridization and methylation studies using MLPA was conducted on 20 patients who were clinically suspected to have PWS. Results and conclusion We analyzed 20 suspected PWS cases descending from 18 unrelated families and 20 healthy controls matching age and sex, using MS-MLPA PWS/AS probemix (MRC-Holland) enables the identification of copy number variations or abnormal methylation patterns. One patient (P18) had PW deletion, which was evident by the reduced copy number ratio, and uniparental disomy was evident in two unrelated patients (P2 and P12). The results indicate that our study identified one typical PWS deletion and two uniparental disomy cases among three patients from distinct families, suggesting that atypical deletions are infrequent in this cohort. This research represents the first exploration of PWS in Egyptian patients using MLPA.

Copy number variations: reliable diagnostic markers for Prader-Willi patients

Background MicroRNAs (miRNAs) are implicated in regulating obesity, but clinical trials have yielded conflicting results. Objective This study aimed to investigate the relationships between circulating levels of miRNA-221 and miRNA-222 in obesity and hypertension-related obesity in humans. This may shed light on the pathogenic pathways controlling obesity and provide noninvasive molecular indicators to identify and predict the disease. The relationships between circulating levels of the above miRNAs and variables related to adiposity and lipid profiles were further investigated. Patients and methods Using a quantitative real-time P technique, the expression levels of circulating miRNAs were determined in serum samples from 65 obese patients (35 without and 30 with hypertension) and 45 age-matched and sex-matched normal-weight individuals. Results and conclusion MiRNAs (221 and 222) have been shown to be differentially expressed in the sera of obese patients compared to controls. In addition, obese hypertensive patients were shown to have higher serum levels of miRNA-222 and miRNA-221 than healthy controls. Serum miRNA-221 was correlated with BMI, cholesterol, triglycerides, and low-density lipoprotein cholesterol. Thus, circulating levels of miRNA-221 and miRNA-222 inease in obesity and obesity-associated hypertension. These miRNAs may serve as markers for obesity and obesity-induced hypertension.

Evaluation of some microRNAs dysregulation in obesity and obesity-related hypertension

Background Obesity is a global epidemic that affects all age groups and social classes. Prevention and treatment strategies against obesity have gained little long-term success. Obesity has been shown to be increased rapidly in the last few decades. It is anticipated that by 2030, up to half of the human population will be classified as obese or overweight. Objective This study aimed to develop a polymeric nanoformulation for the oral delivery of rutin in an obese rat model and to explore the anti-obesogenic impact of this nanosized rutin versus free rutin to provide insightful information for understanding the mechanism of action. Materials and methods Sodium alginate-based nanorutin was prepared and characterized by transmission electron microscope and Zetasizer. The obtained rutin nanoformulation and its free form were applied for obesity management in rats fed a high-fat diet. Results and conclusion The data demonstrated that rutin nanoparticles are made up of single spherical units with a diameter that ranged between 96.1 and 157 nm and exhibited negative zeta potential at −16.6 mV. Treatment of obese rats with chitocal, free rutin, or rutin nanoformulation provoked significant reduction in the body weight, thoracic circumference, BMI, and Lee index. Also, they elicited a significant drop in serum cholesterol, triglycerides, low-density lipoprotein, glucose, insulin, insulin resistance, toll-like receptor 4, nuclear factor kappa B, and leptin levels associated with a significant rise in serum adiponectin and spexin levels. Furthermore, treatment of obese rats with chitocal or rutin nanoformulation elicited significant reduction in body length and abdominal circumference along with significant enhancement in serum high-density lipoprotein level. In conclusion, this approach provides a sparkling proof for the anti-obesogenic potential of rutin nanoformulation through its hypolipidemic, hypoglycemic, and anti-inflammatory effects as well as its inhibitory action on hyperleptinemia and adipogenesis. The superior antiobesity impact of the formulated nanorutin than its free form may be attributed to the enhancement of the solubility and bioavailability by nanoencapsulation.

Background Women often experience significant burdens both at home and in their professional lives, which can lead to physical and mental strain. To manage these challenges, medications such as cyclobenzaprine-HCl, originally developed as a tricyclic antidepressant but reintroduced as a muscle relaxant, are sometimes used. Its effectiveness in treating muscle spasms, localized pain, and limited range of motion in acute musculoskeletal conditions has been well documented. Objective The objective of this study was to explore the potential consequences of cyclobenzaprine-HCl on reproductive health by assessing its influence on female Wistar rats. Materials and methods The study was conducted on 18 adult female Wistar rats, divided into three groups: a control group receiving distilled water and two treated groups with two doses − a low-dose group receiving 1.5 mg/kg of cyclobenzaprine-HCl and a high-dose group receiving 3 mg/kg, administered orally for 30 days. We assessed the impact of cyclobenzaprine-HCl on serum hormonal levels (progesterone, estradiol, luteinizing hormone, follicle-stimulating hormone), oxidative stress markers (glutathione, superoxide dismutase, catalase, malondialdehyde), histopathological changes in ovaries and uteri, and DNA stability using the comet assay. Results and conclusion The study revealed that cyclobenzaprine-HCl caused hormonal imbalances distinguished by significant increase in estradiol and follicle-stimulating hormone levels, and significant decreases in progesterone and luteinizing hormone levels. Dissruptions in oxidative stress markers were characterized by elevated malondialdehyde and reduced glutathione levels. Histopathological abnormalities included degeneration, deformed follicles, congested blood vessels, and necrosis in the ovarian tissue. In addition, diffused eosinophil infiltration, pyknotic nuclei, glandular hyperplasia, necrosis, and hypercellularity in the uterine tissue. DNA instability was observed in both dosage groups as evidenced by fragmented DNA in the shape of comets. These findings underscore the potential reproductive toxicity of cyclobenzaprine-HCl in female rats, suggesting a need for caution in its use considering its possible adverse effects on reproductive health.
