The perception of self and others is a key aspect of social cognition. In order to investigate the neurobiological basis of this distinction we reviewed two classes of task that study self-awareness and awareness of others (theory of mind, ToM). A reliable task to measure self-awareness is the recognition of one's own face in contrast to the recognition of others' faces. False-belief tasks are widely used to identify neural correlates of ToM as a measure of awareness of others. We performed an activation likelihood estimation meta-analysis, using the fMRI literature on self-face recognition and false-belief tasks. The brain areas involved in performing false-belief tasks were the medial prefrontal cortex (MPFC), bilateral temporo-parietal junction, precuneus, and the bilateral middle temporal gyrus. Distinct self-face recognition regions were the right superior temporal gyrus, the right parahippocampal gyrus, the right inferior frontal gyrus/anterior cingulate cortex, and the left inferior parietal lobe. Overlapping brain areas were the superior temporal gyrus, and the more ventral parts of the MPFC. We confirmed that self-recognition in contrast to recognition of others' faces, and awareness of others involves a network that consists of separate, distinct neural pathways, but also includes overlapping regions of higher order prefrontal cortex where these processes may be combined. Insights derived from the neurobiology of disorders such as autism and schizophrenia are consistent with this notion.

Differentiating between self and others: an ALE meta-analysis of fMRI studies of self-recognition and theory of mind.

Functional constipation (FC) is a common functional gastrointestinal disorder (FGID). Neuroimaging studies on patients with FC showed brain functional abnormalities in regions involved in emotional process modulation, somatic and sensory processing and motor control. Brain structural imaging studies in patients with FGID have also shown disease-related alterations in cortical morphometry, but whether and how FC affects brain structure remains unclear. Structural Magnetic Resonance Imaging and surface-based morphometry analysis were used to investigate the impact of FC on cortical morphometry in 29 patients with FC and 29 healthy controls (HC). Results showed that patients with FC compared to HC had significantly decreased cortical thickness in the left middle frontal gyrus (MFG), dorsomedial (DMPFC) and ventromedial prefrontal gyrus (VMPFC), right dorsal anterior cingulate cortex (dACC), left orbitofrontal cortex (OFC), posterior cingulate cortex (PCC)/precuneus, middle temporal gyrus (MTG), and supplementary motor area (SMA) (P < 0.01). Correlation analysis showed that sensation of incomplete evacuation was negatively correlated with cortical thickness in the SMA (P < 0.0001). In addition, patients with FC also had decreased cortical volume than HC in the MTG, precentral gyrus (PreCen) and precuneus/cuneus (P < 0.01), as well as decreased cortical surface area in the PreCen (P < 0.01). No correlation was found between cortical volume/surface area and behavioral measures. These findings suggest that patients with FC are associated with cortical morphometric abnormalities in brain regions implicated in somatic/motor-control, emotional processing and self-referential processing.

Cortical morphometry alterations in brain regions involved in emotional, motor-control and self-referential processing in patients with functional constipation.

Functional constipation (FCon) is a common functional gastrointestinal disorder (FGID); neuroimaging studies have shown brain functional abnormalities in thalamo-cortical regions in patients with FGID. However, association between FCon and topological characteristics of brain networks remains largely unknown. We employed resting-state functional magnetic resonance imaging (RS-fMRI) and graph theory approach to investigate functional brain topological organization in 42 patients with FCon and 41 healthy controls (HC) from perspectives of global, regional and modular levels. Results showed patients with FCon had a significantly lower normalized clustering coefficient and small-worldness, implying decreased brain functional connectivity. Regions showed altered nodal degree and efficiency mainly located in the thalamus, rostral anterior cingulate cortex (rACC), and supplementary motor area (SMA), which are involved in somatic/sensory, emotional processing and motor-control. For the modular analysis, thalamus, rACC and SMA had an aberrant within-module nodal degree and nodal efficiency, and thalamus-related network exhibited abnormal interaction with the limbic network (amygdala and hippocampal gyrus). Nodal degree in the thalamus was negatively correlated with difficulty of defecation, and nodal degree in the rACC was negatively correlated with sensation of incomplete evacuation. These findings indicated that FCon was associated with abnormalities in the thalamo-cortical network.

Cerebral specialization and inter-hemispheric cooperation are two of the most prominent functional architectures of the human brain. Their dysfunctions may be related to pathophysiological changes in patients with Parkinson's disease (PD), who are characterized by unbalanced onset and progression of motor symptoms. This study aimed to characterize the two intrinsic architectures of hemispheric functions in PD using resting-state functional magnetic resonance imaging. Seventy idiopathic PD patients and 70 age-, sex-, and education-matched healthy subjects were recruited. All participants underwent magnetic resonance image scanning and clinical evaluations. The cerebral specialization (Autonomy index, AI) and inter-hemispheric cooperation (Connectivity between Functionally Homotopic voxels, CFH) were calculated and compared between groups. Compared with healthy controls, PD patients showed stronger AI in the left angular gyrus. Specifically, this difference in specialization resulted from increased functional connectivity (FC) of the ipsilateral areas (e.g., the left prefrontal area), and decreased FC in the contralateral area (e.g., the right supramarginal gyrus). Imaging-cognitive correlation analysis indicated that these connectivity were positively related to the score of Montreal Cognitive Assessment in PD patients. CFH between the bilateral sensorimotor regions was significantly decreased in PD patients compared with controls. No significant correlation between CFH and cognitive scores was found in PD patients. This study illustrated a strong leftward specialization but weak inter-hemispheric coordination in PD patients. It provided new insights to further clarify the pathological mechanism of PD.

Brain functional specialization and cooperation in Parkinson’s disease

Major depressive disorder (MDD) is a pervasive mental disorder that significantly impairs functional capabilities, underscoring the necessity for precise stratification of its severity to facilitate tailored treatment. This study investigated the utility of voxel-mirrored homotopic connectivity (VMHC) derived from resting-state functional magnetic resonance imaging (fMRI) data as a neuroimaging biomarker to differentiate varying severities of MDD in a sample drawn from the REST-meta-MDD project, which included 392 first-episode MDD patients and 440 healthy controls (HC) from 9 sites. Patients were classified into mild to moderate and severe depression groups according to the 17-item Hamilton Depression Scale (HAMD) scores. VMHC differences between these subgroups and their associations with HAMD scores were further examined. The results revealed significant reductions in VMHC within the fusiform gyrus for patients with mild to moderate depression compared to HCs, alongside more extensive reductions across the insula, postcentral gyrus, and angular gyrus in severe depression. Notably, increased VMHC in the middle cingulate cortex was identified in severe MDD patients relative to those with mild to moderate depression, with this increase showed a significant positive correlation with the HAMD scores. Additionally, receiver operating characteristic (ROC) curve analysis demonstrated that VMHC values in these regions effectively differentiate patients from HCs and across varying severities of MDD. These findings suggest that VMHC could serve as a valuable metric for clinical diagnosis and the stratification of depression severity, providing insights into the underlying neurobiological mechanisms associated with the disorder.

Influence of depression severity on interhemispheric functional integration: an analysis from the REST-meta-MDD database

Girls who suffer from central precocious puberty (CPP) are at risk of experiencing detrimental psychological and behavioural consequences, along with impaired brain development. However, the mechanism by which puberty hormones affect patients with CPP remains unclear. This study aimed to use degree centrality (DC) analysis to explore the impact of premature activation of the hypothalamic-pituitary-gonadal (HPG) axis on brain functional development in girls with CPP. In this cross-sectional study, thirty-four girls (mean ± SD, 7.89 ± 0.81 years) with CPP and 25 age-matched girls without CPP (mean ± SD, 7.58 ± 0.73 years) underwent resting-state functional magnetic resonance imaging. We used DC analysis to explore brain network properties in CPP girls compared to non-CPP girls. Seed-based FC analysis was performed to identify the connections responsible for the observed differences. Our findings showed that female CPP patients had increased DC in the posterior lobe of the cerebellum and prefrontal areas and increased functional connectivity (FC) between the posterior lobe of the cerebellum and default mode network (DMN) regions relative to age-matched non-CPP girls. Additionally, compared with non-CPP patients, female CPP patients exhibited decreased DC in the bilateral superior parietal gyri and left superior occipital gyrus and reduced FC between the left superior occipital gyrus and right calcarine. A negative correlation was found between basal follicle-stimulating hormone level and DC of the bilateral superior parietal gyri in girls with CPP. The current research provides evidence that premature activation of the HPG axis is associated with the development of cortical function, particularly involving the posterior lobe of the cerebellum, DMN, and prefrontal, parietal, and occipital cortices. Our findings suggest that girls with CPP require attention and early treatment for cognitive and emotional problems as well as brain development in clinical practice.

Altered degree centrality and functional connectivity in girls with central precocious puberty

Autism spectrum disorder (ASD) is a neurodevelopmental disorder accompanied by structural and functional changes in the brain. However, the relationship between brain structure and function in children with ASD remains largely obscure. In the current study, parallel independent component analysis (pICA) was performed to identify inter-modality associations by drawing on information from different modalities. Structural and resting-state functional magnetic resonance imaging data from 105 children with ASD and 102 typically developing children (obtained from the open-access Autism Brain Imaging Data Exchange database) were combined through the pICA framework. Features of structural and functional modalities were represented by the voxel-based morphometry (VBM) and amplitude of low-frequency fluctuations (ALFF), respectively. The relationship between the structural and functional components derived from the pICA was investigated by Pearson's correlation analysis, and between-group differences in these components were analyzed through the two-sample t-test. Finally, multivariate support vector regression analysis was used to analyze the relationship between the structural/functional components and Autism Diagnostic Observation Schedule (ADOS) subscores in the ASD group. This study found a significant association between VBM and ALFF components in ASD. Significant between-group differences were detected in the loading coefficients of the VBM component. Furthermore, the ALFF component loading coefficients predicted the subscores of communication and repetitive stereotypic behaviors of the ADOS. Likewise, the VBM component loading coefficients predicted the ADOS communication subscore in ASD. These findings provide evidence of a link between brain function and structure, yielding new insights into the neural mechanisms of ASD.

Links between brain structure and function in children with autism spectrum disorder by parallel independent component analysis

In recent years, brain signal complexity has gained attention as an indicator of brain well-being and a predictor of disease and dysfunction. Brain entropy quantifies this complexity. Assessment of functional network centrality and connectivity reveals that information communication induces neural signal oscillations in certain brain regions. However, their relationship is uncertain. This work studied brain signal complexity, network centrality, and connectivity in both healthy and depressed individuals. The current work comprised a sample of 124 first-episode drug-naïve patients with major depressive disorder (MDD) and 105 healthy controls (HC). Six functional networks were created for each person using resting-state functional magnetic resonance imaging. For each network, entropy, centrality, and connectivity were computed. Using structural equation modeling, this study examined the associations between brain network entropy, centrality, and connectivity. The findings demonstrated substantial correlations of entropy with both centrality and connectivity in HC and these correlation patterns were disrupted in MDD. Compared to HC, MDD exhibited higher entropy in four networks and demonstrated changes in centralities across all networks. The structural equation modeling showed that network centralities, connectivity, and depression severity had impacts on brain entropy. Nevertheless, no impacts were observed in the opposite directions. This study indicated that the complexity of brain signals was influenced not only by the interactions among different areas of the brain but also by the severity level of depression. These findings enhanced our comprehension of the associations of brain entropy with its influential factors.

The different impacts of functional network centrality and connectivity on the complexity of brain signals in healthy control and first-episode drug-naïve patients with major depressive disorder

The objective of this research was to examine the association between precuneus cortex thickness and mild behavioral impairment (MBI) in patients with mild stroke. Seventy-two patients were evaluated by high-resolution 3 T magnetic resonance and the mild behavioral impairment checklist (MBI-C). To determine the association between precuneus cortex thickness and MBI, we adjusted for demographics, vascular risk factors, and laboratory examination indicators in logistic regression analysis. In addition, we used mendelian randomization to further study the association through genetic databases. Of the 72 mild stroke patients in this study, 26 had MBI. We found a strong negative connection between precuneus cortex thickness and MBI after adjusting for any confounding variables. In patients with an initial mild stroke, the thinner the precuneus cortex, the higher the risk of MBI (OR: 0.02; 95% CI: 0.00-0.39; P < 0.05). Our study has uncovered a significant negative association between the thickness of the precuneus cortex and MBI. This finding provides a novel viewpoint for the radiological diagnosis of MBI, thereby augmenting the contribution of imaging to the diagnostic process of MBI and advancing the prediction of dementia. Specifically, in patients who have suffered mild stroke, a reduction in the cortical thickness of the precuneus has been pinpointed as crucial radiographic evidence of preclinical cognitive impairment. This insight could potentially facilitate earlier detection and intervention strategies for cognitive decline.
