Morphological and phylogenetic studies of Ascomycota from gymnosperms in Sichuan Province, China

Eurotium cristatum is the main probiotic fungus involved in fermentation of Chinese Fuzhuan brick tea, and the amount of E. cristatum is an important indicator of tea quality.Sexual development of E. cristatum consists of three main stages, hypha and ascogonium (at 36 hours post-inoculation), cleistothecium formation (at 48 hours), and cleistothecium ripening (at 72 hours), and each stage exhibits a unique metabolite profile.In this study, developmental changes were analyzed by GC-MS based untargeted metabolomics.Ninety-nine metabolites involved in multiple biochemical pathways were identified.Principal component analysis separated developing E. cristatum into 36, 48 and 72 hour groups.Hierarchical cluster analyses showed that the relative contents of organic acids and amino acids peaked at 36 hours and decreased thereafter, while alcohol and carbohydrate levels peaked at 48-72 hours.Statistical analysis revealed 401 significant metabolite-metabolite correlations (Pearson's |r|>0.8 at a false discovery rate ≤ 0.05), of which 213 were positive and 188 negative.The early increases in organic acid and amino acid metabolites as well as certain phosphate and amine metabolites favor ascogonium formation.Conversely, increased alcohol and carbohydrate concentrations favor cleistothecium formation.These results provide a foundation for further exploration of the biochemical mechanism underlying sexual development in E. cristatum.

Metabolomics reveals changes in metabolite concentrations and correlations during sexual development of Eurotium cristatum (synonym: Aspergillus cristatus)

Colletotrichum is an important plant pathogenic genus that has undergone tremendous taxonomic changes.Species of Colletotrichum also occur as endophytes, saprobes and rarely entomopathogens.The current understanding of Colletotrichum taxonomy and application of various techniques in defining species within the genus is discussed in this paper.Here we provide a review of lifestyles, infection mechanisms, life cycle, host-specificity, classification history and techniques defining Colletotrichum species and the relation to speciation.Misidentifications and mistakes during species introduction are discussed and recommendations are provided for valid species publication.We provide an account of 248 currently accepted species with molecular data, which falls into 14 species-complexes and 13 singleton species.An updated account of Colletotrichum species is provided.Species are listed alphabetically in each species complex and annotated with their habitat, host, geographic distribution, phylogenetic position and typification details.Tables of host specific (152 species) and geographically endemic Colletotrichum species (19 species) are provided.A table of 450 morpho-species is provided for the first time gathering all data to one place, allowing mycologists to check before publishing a new species.Phylogenetic trees are provided for the whole genus and each species complex.Genes and combinations of genes that can be used for the identification of the species complexes are suggested.Future directions for the advancement of this genus are discussed.

Colletotrichum: lifestyles, biology, morpho-species, species complexes and accepted species

Hygrophorus is among the most species rich and frequent ectomycorrhizal genera of Agaricales, and several species are popular wild edible mushrooms in northern and southwestern China.In the past two decades, there has been an increasing number of taxonomic and phylogenetic studies of the genus, however, the comprehensive studies on the species diversity and phylogenetic relationship of Hygrophorus in China were lacking.In this study, an extensive survey on the species diversity of Hygrophorus in China was conducted.In total, 62 Chinese Hygrophorus taxa were included in this study.Fifty-five of them are described, illustrated and discussed, including 26 new species; in addition, two recently described species (H.habaensis and H. viridiflavidus) and 5 insufficiently known species are listed with notes.A large-scale phylogenetic study of Hygrophorus was conducted based on the newly obtained sequences and the available internal transcribed spacer (ITS) sequences in the international public databases; and a multi-gene (ITS-LSU-rpb2-tef1-α) phylogeny was also conducted.The species boundaries of Hygrophorus delimited by morphological data are broadly congruent with those based on the ITS phylogenetic analysis.In general, the multi-locus dataset could resolve a section-level phylogeny of the genus; and a new section, i.e. sect.Vividi, is proposed based on both phylogenetic and morphological evidences.However, the subgenera classified by morphology are not supported well by molecular phylogenetic analysis.
