This study aims to investigate whether the protein isolated from bladder cancer in an Indonesian population can produce the polyclonal antibody for clinical markers of bladder cancer. The participants in this study are bladder cancer patients and healthy persons who were approved for midstream portion urine collection. The inclusion criteria included bladder cancer patients who obtained hematuria examination, urine cytology, and initial therapy with transurethral resection of the bladder; a healthy volunteer who was approved for midstream portion urine collection but without hematuria history, bladder stones, and signs or symptoms of tractus urinarius infection. The procedure consisted of tissue preparation, tissue processing, isolation of membrane cell protein, monitoring of protein in membrane cell, production of polyclonal antibodies, and dot blot technique. A protein with 122 kDa molecular weight is present in epithelial cells of bladder cancer and the normal bladder. A protein with 69 kDa molecular weight is only present in the epithelial cell bladder of normal individuals. In addition, a protein with molecular weight of 47 kDa is only present in epithelial cells of bladder cancer. The minimal ratio of polyclonal antibody with antigen is 1:6400 of the antibody and 1:40 of antigen. Subsequently, this concentration was applied to detect proteins with 47 kDa only in several cancer tissues. Positive results in bladder cancer, but negative results in prostate cancer, rectal cancer, and breast cancer were found. The polyclonal antibody produced from 47 kDa protein subunit is sensitive and specific to detect bladder cancer and become an alternative biomarker for diagnosis and surveillance of bladder cancer.

Polyclonal antibody from 47 kDa protein of bladder cancer is sensitive and specific for detection of bladder cancer

The aim of the present study was to evaluate the in vitro antioxidant activities of the hydro-methanol (2:3) extract of the seeds of Swietenia mahagoni (L.) Jacq. To evaluate the antioxidant activity, the effects of the extract on the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effect, scavenging of hydrogen peroxide, hydroxyl radical scavenging potential, scavenging of nitric oxide, and antilipid peroxidation activity by biochemical methods were examined. Total phenol and flavonoid content in the extract were measured biochemically as per standard methods. Results indicated that the hydro-methanol (2:3) extract had strong scavenging activity on the DPPH radical with an IC50 value 80.54 μg/mL, the hydroxyl radical with an IC50 value 60.76 μg/mL, and hydrogen peroxide with an IC50 value 66.10 μg/mL. The hydro-methanol (2:3) extract also showed notable inhibition of lipid peroxidation with an IC50 value 61.23 μg/mL and nitric oxide with an IC50 value of 81.56 μg/mL. Phytochemical study showed that the extract is rich in phenolic compounds [46.25 mg gallic acid equivalent (GAE)/g dried extract] and flavonoids [231.72 μg quercetin equivalent (QE)/g dried extract]. The present study provides evidence that the hydro-methanol (2:3) extract of seeds of S. mahagoni is a potential source of natural antioxidant activities.

Adiponectin is considered a modulator of glucose metabolism. Hypoadiponectinemia was reported to be associated with glucose metabolism and metabolic syndrome, and had been found in patients with hepatitis B virus (HBV) and hepatitis C virus infections. However, little is known about the role of adiponectin in patients with HBV infection. Fifty-five patients with HBV infection and 52 healthy individuals were included in this study. We found that serum adiponectin was significantly lower in patients with HBV infection. We also found that HBV-infected patients with hemoglobin A1C ≥6.5% had a lower adiponectin level than those with hemoglobin A1C <6.5%. In conclusion, hypoadiponectinemia occurs in patients with HBV infection, and adiponectin may play a role in the pathogenesis of glucose metabolism in these patients.

Serum adiponectin and glucose in patients with hepatitis B virus infection

Prolactin (PRL) and beta-lactoglobulin (BLG) are two important candidate genes well known to be associated with milk production traits as well as somatic cell count (SCC) among dairy cattle breeds. In the present study, the intron 3 of PRL and the spanning region between exon IV and intron IV of the BLG gene were chosen for genotyping and their association with milk production traits as well as SCC among HF crossbred cattle (i.e., Frieswal) that originated from India. We observed that the AA genotype frequency of PRL among Frieswal cows is higher than that of AB and BB. Our findings showed that cows with AA and BB genotypes had significantly (p < 0.05) higher total milk yield and peak yield than AB genotype cows. Comparing SCC with various genotypic groups, we observed that BB genotype cows had significantly (p < 0.05) lower SCC than those with AB and AA genotypes. In the case of BLG, the genotypic frequency of BB was higher than that of AB and AA. The AB and BB genotypes of BLG had a significant (p < 0.05) effect on total milk yield and peak yield compared with AA. The SCC of the AA genotype of BLG is significantly (p < 0.05) lower than that of AB and BB. This study thus indicates that AA and BB genotypes in PRL as well as AB and BB genotypes in BLG may be more suitable for better milk production; however, cows having BB genotype in PRL and AA genotype in BLG may show more resistance to mastitis than those with other genotypes among HF crossbred cattle.

Association of prolactin and beta-lactoglobulin genes with milk production traits and somatic cell count among Indian Frieswal (HF × Sahiwal) cows

Since 1987, cancer has been the leading cause of death in Taiwan. However, chemotherapy resistance in metastatic cancer is the major obstacle to effectively treat cancers. To solve this problem, understanding the mechanism of metastasis is critical. In this study, we used a transwell system to select a metastatic subpopulation from the human lung cancer cell line A549. The invasive and migratory activities of this cell line were analyzed with transwell selection and wound healing assay, respectively. Moreover, migration-stimulating factors, such as SNAI-1 and mPR, were detected with Western blotting. In comparison to A549, enhanced invasiveness, motility, and migration-stimulating factor expression were observed in the invasive lung cancer cell line A549-I5. These cell lines provide a useful tool for the further study of the biologic properties of lung cancer in vitro.

The purpose of the present study was to provide the first evidence of the potential function of the polyherbal combination of soybeans, brown rice, and coconut water (EMSA eritin), as it has been suggested as a therapeutic agent because it, at least in part, increases the production of functional hematopoietic stem cells after irradiation. The study was conducted with 24 male mice (Mus musculus), which were divided into six groups consisting of four mice each. This study used three EMSA eritin doses: 1.04 mg/kg body weight (B.W.), 3.125 mg/kg B.W., and 9.375 mg/kg B.W. All treatment groups, except for normal controls, were exposed to sublethal doses of irradiation and then given EMSA eritin therapy for 15 days. Growth and differentiation of CD4 and CD8 T-lymphocytes in the thymus and the number of stromal cell-derived factor 1 (SDF1)-expressing cells in the spleen, bone marrow, and liver were analyzed using flow cytometry. Results showed that administration of EMSA eritin was capable of significantly inducing lymphocytic proliferation and differentiation and increased the number of SDF1-expressing cells in the spleen, bone marrow, and liver. SDF1 is a chemokine essential to lymphocyte migration and hematopoietic cell development; thus, it can be concluded that EMSA eritin is a good candidate as a therapeutic agent to restore the number of hematopoietic cells after irradiation.

The protective effects of polyherbal extracts of soybeans, coconut water, and brown rice (EMSA eritin) on stromal cell-derived factor 1 production in radiated mice

This study aimed to investigate whether combined supplementation with vitamin C and vitamin E was able to modify the superoxide dismutase (SOD) and malondialdehyde (MDA) levels in the ovarium of rats exposed to rhodamine B. Twenty-five female Wistar albino rats were divided into five groups (n = 5 each), including control (untreated group); rhodamine B group; rhodamine B group which received vitamin C (0.2 mg) + vitamin E (0.04 IU/g body weight); rhodamine B group which received vitamin C (0.4 mg) + vitamin E (0.04 IU/g body weight); and the rhodamine B group which received vitamin C (0.8 mg) + vitamin E (0.04 IU/g body weight). Analysis of MDA levels as a marker of lipid peroxidation was done spectrophotometrically. Analysis of SOD levels was done by enzyme-linked immunosorbent assay technically. Endometrial histology was analyzed in hematoxylin eosin staining. This increase in ovarian MDA was significantly (p < 0.05) attenuated by the two highest dose treatments of combined vitamin C and vitamin E. Rhodamine B significantly decreased SOD levels compared to the untreated group. This decrease in ovarian SOD level was significantly attenuated by the second and third doses of the combined vitamin C and vitamin E. The vascular number and gland density were significantly lower in the rhodamine B group compared to the untreated control group (p > 0.05). All doses also significantly prevented rhodamine B-induced decrease in the vascular number and gland density. In conclusion, the protective effect of combined vitamin C and vitamin E against ovarian and endometrial toxicity in rats receiving oral rhodamine B is due to inhibition of the lipid peroxidation, modulation of SOD levels, and the endometrium repairing effect.
