ACS SENSORS is a peer-reviewed research journal dedicated to disseminating new and original knowledge in all aspects of sensor science related to the selective detection of chemical or biological species or processes. Submitted manuscripts should demonstrate the use of sensors in complex samples, establish their applicability, and relate sensor performance to existing analytical methods. Potentiometric probes used in direct potentiometry are attractive sensing tools. They give information on ion activities, which is often uniquely useful. If, instead, concentrations are desired as sensor output, the ionic strength of the sample must be precisely known, which is often not possible. Here, for the first time, direct potentiometry can be made to report concentrations, rather than activities. It is demonstrated for the detection of monovalent anionic species by using a self-referencing Ag/AgI pulstrode as the reference element instead of a traditional reference electrode. This reference pulstrode releases a discrete quantity of iodide ions from the electrode and the resulting reference potential varies with the activity coefficient of iodide. The effects of activity coefficient on the indicator and reference electrode are therefore compensated and the observed cell potential may now be described in a Nernstian manner against anion concentration, rather than activity. Theoretical simulations and experimental results support the validity of this approach. For most monovalent anions of practical relevance, the potential difference between this approach and from a traditional activity coefficient calculation is less than 0.5 mV. The concept is validated with an all-solid-state nitrate sensor as well as a commercial fluoride-selective electrode, giving Nernstian responses in different ionic strength backgrounds against concentration without the need for correcting activity coefficients or liquid junction potentials.

Direct Potentiometric Sensing of Anion Concentration (Not Activity)

A fundamental limitation of potentiometric ion sensors is their relatively low sensitivity due to the logarithmic dependence between potential and activity. Here we address this issue by exploring a recently developed coulometric transduction method for solid-contact ion-selective electrodes (SCISEs). Spin-coated thin-layer ion-selective membranes are used to lower the membrane resistance and shorten the response time of the SCISEs. When using coulometric transduction, an optimized design of the K+-SCISE is able to detect a concentration change of 5 μM at a concentration level of 5 mM, corresponding to a 0.1% change in K+ activity. This indicates that SCISEs can provide extremely high sensitivity when employing coulometric transduction. Impedance measurements show that the coulometric transduction process for the K+-SCISE is limited by diffusion even for very thin ion-selective membranes. On the other hand, the H+-SCISE shows a low impedance and a fast coulometric response that is related to the high mobility of H+ in the H+-selective polymeric membrane as well as in the solid contact layer. The coulometric transduction method was used to detect small changes of pH in seawater and found to improve the sensitivity compared to classical potentiometry. The coulometric method was briefly tested also for determining activity changes of K+ in a serum sample.

Improving the Sensitivity of Solid-Contact Ion-Selective Electrodes by Using Coulometric Signal Transduction

Rapid and simple nucleic acid detection is significant for disease diagnosis and pathogen screening, especially under specific conditions. However, achieving highly sensitive and specific nucleic acid detection to meet the time and equipment demand remains technologically challenging. In this study, we proposed a magnetic separation enhanced colorimetry biosensor based on a toehold-containing three-way junction (TWJ) induced multiple isothermal exponential amplification and the CRISPR/Cas14a (C-TEC) biosensor. The TWJ template was designed as a Y–X–Y structure. In the presence of the target, the formation of toehold-containing TWJ complex induced primer extension, leading to the generation of amplified single-stranded DNA; this amplified DNA could then bind to either the free TWJ template for EXPAR reaction or the toehold of the TWJ complex for toehold-mediated strand displacement, thereby enabling the recycling of the target. The amplification products could trigger CRISPR/Cas14a for efficient trans-cleavage and release the magnetically bound gold nanoparticle probes for colorimetry detection. Using Mycobacterium tuberculosis 16S rDNA as the target, the proposed C-TEC could detect 16S rDNA down to 50 fM by the naked eye and 20.71 fM by UV–vis detector at 520 nm within 90 min under optimal conditions. We successfully applied this biosensor to clinical isolates of Mycobacterium tuberculosis. In addition, the C-TEC biosensor also showed feasibility for the detection of RNA viruses. In conclusion, the proposed C-TEC is a convenient, fast, and versatile platform for visual detection of pathogen DNA/RNA and has potential clinical applications.

Toehold-Containing Three-Way Junction-Initiated Multiple Exponential Amplification and CRISPR/Cas14a Assistant Magnetic Separation Enhanced Visual Detection of <i>Mycobacterium Tuberculosis</i>

A portable and highly reproducible lab-on-capillary surface-enhanced Raman scattering (SERS) platform was developed using a specially designed homemade device for rapid on-site SERS measurement. In particular, this platform was composed of a capillary with a tiny orifice, which allows an effective and lossless sample extraction, resulting in high SERS performance. The capillary-based plasmonic substrate was prepared by compactly assembling Au@Ag core–shell nanorods (NRs) embedded with the 4-mercaptobenzoic acid (4-MBA) molecule as an internal standard onto the inner wall of a capillary tube. The fabrication process is facile and convenient with no requirement for complicated procedures. The exclusively prepared nanoparticles were able to significantly improve the signal consistency and overcome the limitations of reliable quantitative SERS analysis compared with conventional methods. Importantly, it was found that this capillary-based substrate with higher sensitivity was essentially attributed to more valid nanoparticles in the effective laser excitation region derived from the unique structure of the capillary. Furthermore, the applicability of the Au@4-MBA@Ag nanorod-decorated capillary for the quantitative identification of fungicides (malachite green and crystal violet) on the shell was demonstrated. As a result, this proposed lab-on-capillary sensor holds promising practical potential for rapid on-site analysis, especially for various contaminants on an uneven surface.

Lab-On-Capillary Platform for On-Site Quantitative SERS Analysis of Surface Contaminants Based on Au@4-MBA@Ag Core–Shell Nanorods

The "turn-on" mode surface-enhanced Raman scattering (SERS) aptasensor for ultrasensitive ochratoxin A (OTA) detection was developed based on the SERS "hot spots" of AuNanostar@4-MBA@Au core–shell nanostructures (AuNS@4-MBA@Au) and exonuclease III (Exo III)-assisted target cycle amplification strategy. Compared with conventional gold nanoparticles, AuNS@4-MBA@Au provides a much higher SERS enhancement factor because AuNS exhibits a larger surface roughness and the lightning rod effect, as well as an excellent electromagnetic field between the AuNS core and the Au shell, which contribute to the superstrong SERS signal. Meanwhile, Exo III-assisted target cycle amplification can be used as an effective method for the further amplified detection of OTA. Additionally, the utilization of streptavidin magnesphere paramagnetic particles offers a green, economical, and facile technology for the accumulation and separation of the signal probe AuNS@4-MBA@Au from solution. All these factors lead to a significant enhancement of detectable signals and superhigh sensitivity. As a result, the limit of detection as low as 0.25 fg mL–1 could be achieved, which was lower than that in the other reported literatures on SERS methods for OTA detection as we know. The developed SERS aptasensor also provides a promising tool for foodstuff detection.

Humidity sensors were widely used in Internet of Things (IoT) applications. According to medical respiration monitoring, noncontact sensing, and human-computer interface requirements, fast response and high repeatability are important for efficient and precise signal acquisition. Previous research works mainly focus on sensitivity improvement with the scarification of response time, stability, and repeatability. In this paper, we proposed the surface acoustic wave (SAW) humidity sensors with comprehensive performance using the uniform multiwrinkled graphene oxide (GO) films as the sensing material obtained by vacuum filtration and liquid phase transfer method. The multiwrinkled GO films offered controllable thickness (thin to 29 nm), uniform wafer-level fabrication (2 in.), and abundant wrinkle, making them become effective sensitive films for SAW humidity sensors due to numerous adsorption sites and transfer channels for water molecules. The experimental results showed that the sensors can obtain high sensitivity (10.5 kHz/RH%@60 nm thick film), ultrafast response (∼45 ms), good stability (variation amplitude ∼0.1%), repeatability (variation amplitude ∼1%), and wafer-level fabrication capability demonstrating their practical applications for medical respiration monitoring, noncontact sensing, and human-computer interface.

A High-Performance Surface Acoustic Wave Humidity Sensor with Uniform Multiwrinkled Graphene Oxide Film

The frequent and excessive use of organophosphorus pesticides in the agriculture industry raises persistent concerns regarding their environmental protection and public health implications. Addressing these issues requires the development of affordable and reliable sensing platforms for on-field monitoring to mitigate their adverse impacts promptly. This study utilizes nanocellulose papers (bacterial and TEMPO-oxidized) combined with butyrylcholinesterase to create a novel reagent-free and orthogonal nanobioplatform featuring smart opto-electrochemical dual outputs. An integrated nano-PAD, preloaded with enzymes and enzymatic substrates, is fabricated using wax-printing and screen-printing technologies. The nano-PAD measures opto-electroactive products, specifically indoxyl and thiocholine, whose concentrations correlate directly with the enzymatic inhibition caused by paraoxon, used as the organophosphate model. To enhance user convenience and meet the requirements for smart real-time point-of-need detection, integration of the nano-PAD with a smartphone-operated miniaturized potentiostat and a self-developed portable smart optical reader is achieved. The developed bioanalytical platform, further supported by a self-developed Android application, enables accurate and efficient quantification of dual signals in real time. The system covers a wide detection range of paraoxon (20-100 ppb) and demonstrates reliable recovery levels (ranging from 98 to 107%) in a real matrix, specifically wastewater. Given these demonstrated capabilities, this innovative biosensing strategy holds substantial potential for practical application in environmental surveillance, facilitating timely and informed environmental management decisions, particularly in resource-limited settings where traditional analytical tools are inaccessible.

Nanopaper Integrated Smart Device: An Opto-Electrochemical Biosensor for Real-Time Dual On-Field Detection of Organophosphorus Pesticides

Eyelid pressure is a crucial biomechanical parameter for ocular health and refractive status, yet measuring it poses challenges related to flexibility, sensitivity, and regional specificity. This study introduces a novel smart contact lens that incorporates kirigami designs and an iontronic capacitive sensing array to enhance flexibility and conformability. The unique structural composition of this device allows for precise and simultaneous monitoring of eyelid pressure in multiple regions with a high sensitivity and seamlessly fit across corneal curvatures. The efficacy of the sensor has been thoroughly confirmed through comprehensive evaluations in rabbits and porcine eyes, demonstrating improved conformity and sensitivity compared to conventional single-point sensors. Assessments have been conducted in various conditions, including under anesthesia and in awake states, as well as the deliberate alteration of intraocular pressure fluctuations, all affirming the exceptional accuracy in detecting eyelid pressure. We envision that the smart contact lens has the potential to revolutionize diagnosis and management of eyelid-related ocular diseases.

Kirigami Design Smart Contact Lens for Highly Sensitive Eyelid Pressure Measurement

Fe single-atom and Fe cluster-coupled N, S co-doped carbon nanomaterials (FeSA–FeONC-NSC) were prepared through a two-step high-temperature pyrolysis process using Gelidium corneum enriched with C, Fe, O, N, and S as precursors. The analysis by aberration-corrected scanning transmission electron microscopy and X-ray absorption spectroscopy revealed the presence of single-atom Fe in Fe–N4 coordination structures, along with small clusters as Fe–O-coordinated Fe2O3. Single-atom Fe in the form of Fe2+/Fe3+ provides more electrocatalytic active sites, which synergistically accelerates the charge migration process in the assembly of FeSA–FeONC-NSC with Fe2O3 clusters. The flexible nonenzymatic sensor, based on FeSA–FeONC-NSC and fabricated using a polydimethylsiloxane substrate, exhibited excellent catalytic activity for both uric acid (UA) and tyrosine (Tyr). Low detection limits for UA (0.14 μmol L–1) and Tyr (0.03 μmol L–1) were observed by using chronoamperometry in artificial sweat. The in situ detection of sweat was performed in combination with an integrated circuit board affixed to human skin, and the results were generally consistent with those of the high-performance liquid chromatography method. Therefore, FeSA–FeONC-NSC serves as a good modifier for wearable electrochemical sweat sensor applications.
