Abstract The internal organs embedded in the cavities are lined by an epithelial monolayer termed the mesothelium. The mesothelium is increasingly implicated in driving various internal organ pathologies, as many of the normal embryonic developmental pathways acting in mesothelial cells, such as those regulating epithelial-to-mesenchymal transition, also drive disease progression in adult life. Here, we summarize observations from different animal models and organ systems that collectively point toward a central role of epithelial-to-mesenchymal transition in driving tissue fibrosis, acute scarring, and cancer metastasis. Thus, drugs targeting pathways of mesothelium’s transition may have broad therapeutic benefits in patients suffering from these diseases.

Abstract The integrated stress response (ISR) plays a pivotal role in the cellular stress response, primarily through global translational arrest and the upregulation of cellular adaptation-linked molecules. Growth differentiation factor 15 (Gdf15) is a potent stress-responsive biomarker of clinical inflammatory and metabolic distress in various types of diseases. Herein, we assess whether ISR-driven cellular stress contributes to pathophysiological outcomes by modulating Gdf15. Clinical transcriptome analysis demonstrates that PKR is positively associated with Gdf15 expression in patients with renal injury. Gdf15 expression is dependent on protein kinase R (PKR)-linked ISR during acute renointestinal distress in mice and genetic ablation of Gdf15 aggravates chemical-induced lesions in renal tissues and the gut barrier. An in-depth evaluation of the gut microbiota indicates that Gdf15 is associated with the abundance of mucin metabolism-linked bacteria and their enzymes. Moreover, stress-responsive Gdf15 facilitates mucin production and cellular survival via the reorganization of the autophagy regulatory network. Collectively, ISR-activated Gdf15 counteracts pathological processes via the protective reprogramming of the autophagic network and microbial community, thereby providing robust predictive biomarkers and interventions against renointestinal distress.

Stress-responsive Gdf15 counteracts renointestinal toxicity via autophagic and microbiota reprogramming

Plant functional traits (FTs) determine growth, reproduction and survival strategies of plants adapted to their growth environment. Exploring global geographic patterns of FTs, their covariation and their relationships to climate are necessary steps towards better-founded predictions of how global environmental change will affect ecosystem composition. We compile an extensive global dataset for 16 FTs and characterise trait-trait and trait-climate relationships separately within non-woody, woody deciduous and woody evergreen plant groups, using multivariate analysis and generalised additive models (GAMs). Among the six major FTs considered, two dominant trait dimensions-representing plant size and the leaf economics spectrum (LES) respectively-are identified within all three groups. Size traits (plant height, diaspore mass) however are generally higher in warmer climates, while LES traits (leaf mass and nitrogen per area) are higher in drier climates. Larger leaves are associated principally with warmer winters in woody evergreens, but with wetter climates in non-woody plants. GAM-simulated global patterns for all 16 FTs explain up to three-quarters of global trait variation. Global maps obtained by upscaling GAMs are broadly in agreement with iNaturalist citizen-science FT data. This analysis contributes to the foundations for global trait-based ecosystem modelling by demonstrating universal relationships between FTs and climate.

Global patterns of plant functional traits and their relationships to climate.

Abstract Early studies of the free-living nematode C. elegans informed us how BCL-2-regulated apoptosis in humans is regulated. However, subsequent studies showed C. elegans apoptosis has several unique features compared with human apoptosis. To date, there has been no detailed analysis of apoptosis regulators in nematodes other than C. elegans . Here, we discovered BCL-2 orthologues in 89 free-living and parasitic nematode taxa representing four evolutionary clades (I, III, IV and V). Unlike in C. elegans , 15 species possess multiple (two to five) BCL-2-like proteins, and some do not have any recognisable BCL-2 sequences. Functional studies provided no evidence that BAX/BAK proteins have evolved in nematodes, and structural studies of a BCL-2 protein from the basal clade I revealed it lacks a functionally important feature of the C. elegans orthologue. Clade I CED-4/APAF-1 proteins also possess WD40-repeat sequences associated with apoptosome assembly, not present in C. elegans , or other nematode taxa studied.

Diversity in the intrinsic apoptosis pathway of nematodes

Age at menarche (AAM) and age at natural menopause (ANM) are highly heritable traits and have been linked to various health outcomes. We aimed to identify circulating proteins associated with altered ANM and AAM using an unbiased two-sample Mendelian randomization (MR) and colocalization approach. By testing causal effects of 1,271 proteins on AAM, we identified 22 proteins causally associated with AAM in MR, among which 13 proteins (GCKR, FOXO3, SEMA3G, PATE4, AZGP1, NEGR1, LHB, DLK1, ANXA2, YWHAB, DNAJB12, RMDN1 and HPGDS) colocalized. Among 1,349 proteins tested for causal association with ANM using MR, we identified 19 causal proteins among which 7 proteins (CPNE1, TYMP, DNER, ADAMTS13, LCT, ARL and PLXNA1) colocalized. Follow-up pathway and gene enrichment analyses demonstrated links between AAM-related proteins and obesity and diabetes, and between AAM and ANM-related proteins and various types of cancer. In conclusion, we identified proteomic signatures of reproductive ageing in women, highlighting biological processes at both ends of the reproductive lifespan.

Mendelian randomization identifies circulating proteins as biomarkers for age at menarche and age at natural menopause

Abstract Eye tracking has emerged as a key method for understanding how animals process visual information, identifying crucial elements of perception and attention. Traditional fish eye tracking often alters animal behavior due to invasive techniques, while non-invasive methods are limited to either 2D tracking or restricting animals after training. Our study introduces a non-invasive technique for tracking and reconstructing the retinal view of free-swimming fish in a large 3D arena without behavioral training. Using 3D fish bodymeshes reconstructed by DeepShapeKit, our method integrates multiple camera angles, deep learning for 3D fish posture reconstruction, perspective transformation, and eye tracking. We evaluated our approach using data from two fish swimming in a flow tank, captured from two perpendicular viewpoints, and validated its accuracy using human-labeled and synthesized ground truth data. Our analysis of eye movements and retinal view reconstruction within leader-follower schooling behavior reveals that fish exhibit negatively synchronised eye movements and focus on neighbors centered in the retinal view. These findings are consistent with previous studies on schooling fish, providing a further, indirect, validation of our method. Our approach offers new insights into animal attention in naturalistic settings and potentially has broader implications for studying collective behavior and advancing swarm robotics.

Non-invasive eye tracking and retinal view reconstruction in free swimming schooling fish

Abstract GABAergic synapses critically modulate neuronal excitability, and plastic changes in inhibitory synaptic strength require reversible interactions between GABA A receptors (GABA A Rs) and their postsynaptic anchor gephyrin. Inhibitory long-term potentiation (LTP) depends on the postsynaptic recruitment of gephyrin and GABA A Rs, whereas the neurotransmitter GABA can induce synaptic removal of GABA A Rs. However, the mechanisms and players underlying plastic adaptation of synaptic strength are incompletely understood. Here we show that neurobeachin (Nbea), a receptor trafficking protein, is a component of inhibitory synapses, interacts with gephyrin and regulates the downscaling of inhibitory synaptic transmission. We found that the recruitment of Nbea to GABAergic synapses is activity-dependent and that Nbea regulates GABA A R internalization in a protein kinase A (PKA)-dependent manner. In heterozygous neurons lacking one Nbea allele, re-expression of Nbea but not expression of a PKA binding-deficient Nbea mutant rescued the internalization of GABA A Rs. Our data suggest a mechanism by which Nbea mediates PKA anchoring at inhibitory postsynaptic sites to downregulate GABAergic transmission. They emphasize the importance of kinase positioning in the regulation of synaptic strength.

Abstract The microbial dimension of the terroir is crucial for wine quality, as microbiomes contribute to plant biofertilization, stress tolerance and pathogen suppression. While microbial terroir can act as a biological signature at large scale, data for local contexts is lacking, hindering the characterization of regional microbial diversity in vineyards. Here, we define the microbial terroir of vineyards across the 12 sub-areas (Additional Geographic Units -AGUs) of the “Consorzio del Vino Nobile di Montepulciano DOCG” PDO area (Italy), a world-renowned wine-producing region. Rhizospheres of Vitis vinifera cultivar Sangiovese and soil samples were collected throughout the 2022 viticultural season and analyzed through an integrated metabarcoding/shotgun metagenomic approach, targeting bacteria and fungi. Wine metabolomics was also perfomed, projecting compositional and functional variations of the microbial terroir at the AGUs level into a corresponding variation in the product metabolic profile. Our findings reveal a unique taxonomic configuration of the Vino Nobile di Montepulciano terroir compared to other vineyards, with microbiomes being “AGU-specific” in taxonomic abundances and plant growth-promoting functions, confirming the potential relevance of characterizing and preserving the microbial terroir to safeguard high-quality traditional wines.
