Introduction: Despite high rates of mortality and morbidity due to preterm delivery, none of the diagnostic and biochemical tests could help to accurately identify the cases at risk of preterm delivery. Objectives: Since ferritin is an acute phase reactant, this study aimed to evaluate serum ferritin levels in women with preterm and term delivery. Methods: This study was conducted on 150 women with preterm delivery (50 cases in each of the groups 24-30, 30-34, and 34-37 weeks of pregnancy), 150 women with term delivery, and 150 pregnant women who were between 24-37 weeks of pregnancy (same as preterm delivery groups). Results: The mean ferritin level in all preterm groups was significantly higher than that in the term group, but there was no difference among the preterm groups. In addition, ferritin level in each preterm group was significantly higher than those in the normal pregnancy group at the same gestational age. In preterm deliveries, ferritin levels were significantly higher in cases with prelabor rupture of membranes (PROM) or with a prolonged leakage (longer than 12 hours). In addition, in patients with PROM or prolonged leakage, ferritin levels were significantly higher in preterm deliveries than in term deliveries. A ferritin level of 37.5 ng/mL recognized as the best cut-off for preterm delivery, as compared with term delivery, and its sensitivity, specificity, and diagnostic accuracy was 78.7%, 68.7%, and 73.6%, respectively. Conclusion: The findings of the present study showed that serum ferritin level can be used to find patients at risk of preterm delivery.

Introduction: The prevalence of Janus kinase 2 V617F, Jak 2-Exon12, calreticulin (CALR) gene mutations and myeloproliferative leukemia mutations in myeloproliferative neoplasms (MPNs) without BCR-ABL1 gene fusion rearrangement in Iraq remains unknown due to inadequate data. Objectives: The aim of this study was to ascertain the prevalence of these mutations and assess the clinical characteristics of polycythemia vera (PV), essential thrombocythemia (ET) and primary myelofibrosis (PMF) in Erbil, Iraq. Patients and Methods: In this cross-sectional study, 80 patients were evaluated for the presence of JAK 2 V617, CALR, and MPL mutations. Specific primers were utilized to amplify MPNs-associated mutations (JAK 2 V617F, Jak 2 -Exon12, CALR, and MPL) using DNA extracted from whole blood. Results: The presence of the JAK 2 V617F mutation was identified in 96% of patients diagnosed with PV and 35% of patients diagnosed with ET. Additionally, JAK 2 exon 12 was detected in 4% of PV patients. CALR mutation was identified in 65% of ET patients. No detection of MPL mutation was observed. A higher prevalence of the CALR mutation was observed in younger age groups, whereas the JAK 2 V617F mutation was predominantly found in older age groups. Conclusion: The incorporation of genetic tests for JAK 2 V617F, JAK 2 exon 12, MPL exon 10, and CALR exon 9 significantly improves the diagnostic effectiveness for BCR ABL1-gene fusion negative MPNs.

Evaluation of Philadelphia-negative myeloproliferative neoplasms in Erbil Iraq; analysis of JAK 2, CALR, and MPL mutations

Introduction: SARS-CoV-2 was a new strain of the Coronaviridae family. It caused the coronavirus disease in December 2019 that first arose in Wuhan, China, and it developed into a worldwide health issue. COVID-19 gives rise to a wide range of symptoms which can range from having no symptoms to having extensive fatal pneumonia. Objectives: The purpose of the study was to figure out how infecting the virus could influence and impact the various laboratory biomarkers among dead male and female patients. Patients and Methods: The data and statistics were obtained from the dead patients of West Emergency Hospital and the data was investigated directly. The investigation was conducted from June 1st to November 1st, 2020. Epidemiological, clinical as well as lab data of approximately 659 dead cases of SARS-CoV-2 infected individuals were examined in that period. A real-time polymerase chain reaction (RT-PCR) technique was applied to confirm infecting of SARS-CoV-2. The deceased cases were categorized into 2 fractions, which were a male and female group. The male group had 407 individuals and the female group had 252 individuals. Results: This study revealed that the male cases had lower lymphocyte and platelet counts than females but substantially higher levels of urea and creatinine, also the male cases were older compared to the female cases. However, D-dimer and C-reactive protein (CRP ) levels did not indicate any notable differences between the two groups. Conclusion: The study showed a gender-situated differences in the laboratory biomarkers of dead COVID-19 cases.

Deciphering gender disparities in laboratory biomarkers among deceased COVID-19 patients in Erbil city-Iraq: A retrospective study

Introduction: A systematic library of Gal4/UAS-regulated transgenes has proven to be a powerful genetic system for identifying genes and defining pathways in development. This system offers valuable insights that highlight the evolutionary conservation between animals and humans. Objectives: The objective of this study was to clone, express, and characterize the UbiA gene. The research presents a highly effective method for cloning genes, using the UbiA-pcDNA3 gene as a model for mammalian cloning. These genes were then integrated into the PUAST vector of Drosophila, an expression vector and eukaryotic cell system commonly used for producing recombinant proteins. Materials and Methods: UbiA was isolated from human cells, and complementary DNA was synthesized. An oligonucleotide primer pair was designed based on the UbiA gene sequence, incorporating XhoI and Xbal restriction sites at the 5´ end of the forward and reverse primers, respectively. The UbiA gene was then amplified by PCR, cloned into the pcDNA3 plasmid, and the resulting recombinant plasmid was sequenced. Subsequently, the gene was sub cloned into the PUAST vector and expressed in S2 cells as a eukaryotic cell system. Protein determination and verification were conducted through western blotting techniques. Results: Confirmation of UbiA gene cloning into the PUAST vector was achieved through colony-PCR and digestion by enzymes. Cloning and sub cloning techniques validated by enzymatic digestion, along with gene sequencing. The identity between cloned UbiA gene and the identical gene exhibited 99%. We revealed a singular band purified protein through western blotting with 60 kDa size. Conclusion: More protein synthesis of the UbiA gene can be achieved by using the eukaryotic expression system provided by the PUAST vector. This technique has been proven to be a suitable platform and can be instrumental in various applications such as therapeutics, pharmacology, and vaccine development.

Cloning and expression of UbiA human gene using innovative methodologies for recombinant protein production in PUAST vector

Introduction: Osteoarthritis is an inflammatory disorder and the most common human joint disease. Considering the anti-inflammatory properties of selenium, we aimed to investigate the relationship between selenium and the risk of osteoarthritis in this study. Materials and Methods: The present study was a systematic review and meta-analysis based on the PRISMA guidelines, conducted in databases including Web of Science, Cochrane, ProQuest, PubMed, and Google Scholar Search Engine for articles published until December 12, 2023. Data was analyzed using STATA 14 software, and tests with p-values lower than 0.05 were considered statistically significant. Results: Seven cross-sectional studies with 412998 participants were combined, and results indicated that selenium intake increased the risk of osteoarthritis (OR: 1.26; 95% CI: 1.06, 1.49). However, the risk of osteoarthritis in individuals with low, moderate, and high serum selenium levels was (OR: 1.03; 95% CI: 0.81, 1.31), (OR: 0.97; 95% CI: 0.83, 1.13), and (OR: 0.99; 95% CI: 0.99, 1), respectively. The relationship between serum selenium level and the risk of osteoarthritis in male and female patients were also (OR: 0.99; 95% CI: 0.98, 0.99) and (OR: 0.99; 95% CI: 0.98, 0.99), respectively. Findings showed the relationship between serum selenium level and knee (OR: 1.11; 95% CI: 0.89, 1.38) and hip (OR: 1.23; 95% CI: 0.86, 1.77) osteoarthritis. Conclusion: Selenium intake increased the risk of osteoarthritis by 26%. However, there was no statistically significant relationship between the serum selenium level and the risk of osteoarthritis. Considering the limited number of reviewed studies, further studies in this field are required. A systematic review and meta-analysis of the association between selenium and osteoarthritis

Introduction: Freezing of gait (FOG) is a debilitating symptom experienced by many individuals with Parkinson’s disease, characterized by an abrupt and transient inability to initiate or continue locomotion. Despite extensive research, effective pharmacological interventions targeting FOG remain elusive. Atomoxetine, a selective norepinephrine reuptake inhibitor, has shown promise in alleviating FOG in small-scale studies. However, the efficacy of atomoxetine in larger cohorts remains underexplored. Objectives: This study aims to assess the impact of atomoxetine on Parkinson’s disease-related FOG in a larger study population than currently reported in the literature. Patients and Methods: This randomized clinical trial was performed in Golestan Hospital, Ahvaz, Iran, in 2022. The participants were divided into group A comprising 16 patients subjected to atomoxetine treatment, and group B consisting of 16 patients administered a placebo. Three scheduled visits were conducted. Freezing of Gait Questionnaire (FOGQ) scores was documented during each visit. Results: The mean age of all participants was 62.4 ± 8.3 years, with no significant intergroup age disparity and no statistically significant gender-based distinctions between the two groups. The average disease duration in the atomoxetine group was 8.2 ± 3.1 years, while in the placebo group, it was 7.2 ± 3.4 years, with no significant divergence between the two groups. The study findings indicated no significant distinction in FOGQ scores between the treatment and placebo groups. Conclusion: Though atomoxetine did not significantly improve improvement in freezing of the gait of studied patients with Parkinson’s disease, it may cause a modest improvement in the gait freezing of some patients. Trial Registration: The trial protocol was approved by the Iranian Registry of Clinical Trials (identifier: IRCT20211008052695N1; https://irct.behdasht.gov.ir/trial/59643, ethical code; #IR.AJUMS.HGOLESTAN. The effect of atomoxetine efficacy on freezing of gait in patient with Parkinson’s disease: a double-blinded randomized clinical trial

Introduction: Colorectal malignancy is the third furthermost frequent malignancy in the world, and approximately 80% of the cases are related to nongenetic causes, including high cholesterol levels. Accordingly, the present systematic review and meta-analysis aimed to investigate the relationship between statin treatment and the risk of colorectal cancer. Materials and Methods: The current study was a systematic review and meta-analysis designed based on PRISMA. International databases, including PubMed, Scopus, Web of Science, Cochrane, and Google Scholar search engine, were used to search for studies published until July 7, 2023. Data analysis was directed by STATA software version 14, and P values below than 0.05 (P&lt;0.05) indicated the significance of statistical tests. Results: The results obtained from a combination of 30 studies with 2436650 samples indicated that statin use increased the risk of colorectal malignancy by 11% (OR: 0.89; 95% CI: 0.84, 0.95) and reduced the risk of rectal cancer by 16% (R: 0.84; 95% CI: 0.74, 0.94). Instead, statin administration reduced the hazard of colorectal cancer in individuals aged 50 to 59 by 22% (R: 0.78; 95% CI: 0.63, 0.95), 60 to 69 by 14% (OR: 0.86; 95% CI: 0.75, 0.98), and 70 to 79 by 8% (OR: 0.92; 95% CI: 0.87, 0.98). Utilization of statin in male (OR: 0.71; 95% CI: 0.58, 0.87) and female (OR: 0.84; 95% CI: 0.77, 0.92) patients reduced the risk of colorectal cancer by 29% and 16%, respectively. Furthermore, statin use in men (OR: 0.41; 95% CI: 0.26, 0.67) and women (OR: 0.64; 95% CI: 0.42, 0.97) reduced the risk of colon cancer by 59% and 36%, respectively. Conclusion: Statin administration reduced the risk of colorectal cancer, and the possibility of reduction in risk of colorectal cancer at lower ages was higher than in older adults and also higher in males compared with females. Statins and the risk of colorectal cancer; a systematic review and meta-analysis of cohort and case-control studies

Introduction: Cholelithiasis and related treatments are associated with several gastrointestinal cancers. Thus, the present systematic review and meta-analysis aimed to investigate the relationship between cholelithiasis and cholecystectomy with the risk of liver cancer. Materials and Methods: The present study is a systematic review and meta-analysis conducted using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA). We performed a search on the databases of Web of Science, Cochrane, ProQuest, PubMed, and Google Scholar until January 21, 2024. Moreover, the statistical analysis was performed using the STATA 14, and the significance level was set at P&lt;0.05. Results: The present meta-analysis included 15 studies with a total sample size of 1426704 participants. According to our results, a significant association between cholelithiasis and liver cancer (OR: 1.76, 95% CI: 1.52, 2.03), as well as cholecystectomy and liver cancer (OR: 1.55, 95% CI: 1.29, 1.87) were detected. Moreover, the relationship between cholelithiasis and liver cancer was significant in cohort studies (OR: 1.76, 95% CI: 1.51, 2.04) and insignificant in case-control studies (OR: 1.69, 95% CI: 0.86). However, there was a significant relationship between cholecystectomy and liver cancer in both the cohort (OR: 1.62, 95% CI: 1.29, 2.04) and case-control studies (OR: 1.26, 95% CI: 1.12, 1.41). Conclusion: In conclusion, cholelithiasis and cholecystectomy increased the risk of liver cancer by 76% and 55%, respectively. Thus, they can be considered as risk factors for liver cancer. Systematic review and meta-analysis of the association between gallstones and cholecystectomy with liver cancer

Introduction: Breast cancer (BC) is among the top causes of mortality among women worldwide. Identifying genes by differential expression associated with the development of the disease helps us to better understanding the molecular mechanisms of BC. Objectives: Our study used in-silico analysis to identify hub genes could trigger the development of BC. Materials and Methods: We identified GSE38959 and GSE45827 for differentially expressed genes (DEGs) in the Gene Expression Omnibus (GEO) database, with an adjusted P&lt;0.05. In both sets, logFC ≥ 2 and logFC ≤ -2 were observed in the DEGs that express themselves within cases and normal BC samples. A comparison was then performed, detecting two common datasets of DEGs using the GEO2R tool. Pathways were elucidated using the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology databases. Thereafter, protein-protein interactions (PPIs) were analyzed using Cytoscape and Gephi. Finally, a GEPIA analysis was conducted to validate the target genes. Results: Using the GEO, 322 common DEGs were identified and 65 hub genes as PPIs. The DEGs were enriched in functions associated with cell division, chromosomes, centromeric regions, microtubule binding, and the cell cycle based on the gene ontology (GO) and KEGG pathways analysis. The expression of 6 genes, CDK1, CCNB1, TOP2A, CXCL12, IGF1, and KIT, represented statistically significant values when the normal and tumor samples were compared via GEPIA analysis. Conclusion: This study introduced six genes (CDK1, CCNB1, TOP2A, CXCL12, IGF1, and KIT) with high expression significantly, which could act as a biomarker for BC development (P&lt;0.05 for all genes). Further comprehensive experimental in vivo studies are needed to describe their role in BC.
