A synergistic therapy agent (STA) with photothermal, photodynamic, chemodynamic, and starvation therapy (PTT, PDT, CDT, and ST) functions was developed. Hollow, mesoporous, and nearly uniform CeO2 nanoparticles (H-CeO2 NPs) were synthesized using a staged shape templating sol–gel protocol. Chlorin e6 (Ce6) was adsorbed onto H-CeO2 NPs, and a thin polydopamine (PDA) layer was formed on Ce6-adsorbed H-CeO2 NPs. Glucose oxidase (GOx) was bound onto PDA-coated Ce6-adsorbed H-CeO2 NPs to obtain the targeted STA (H-CeO2@Ce6@PDA@GOx NPs). A reversible photothermal conversion behavior with the temperature elevations up to 34 °C was observed by NIR laser irradiation at 808 nm. A cascade enzyme system based on immobilized GOx and intrinsic catalase-like activity of H-CeO2 NPs was rendered on STA for enhancing the effectiveness of PDT by elevation of ROS generation and alleviation of hypoxia in a tumor microenvironment. Glucose-mediated generation of highly toxic hydroxyl radicals (·OH) was evaluated for CDT. The effectiveness of PDT on glioblastoma T98G cells was markedly enhanced by O2 generation started by the decomposition of glucose. A similar increase in cell death was also observed when ST and CDT functions were enhanced by photothermal action. The viability of T98G cells decreased to 10.6% by in vitro synergistic action including ST, CDT, PDT, and PTT without using any antitumor agent.

In Vitro Synergistic Photodynamic, Photothermal, Chemodynamic, and Starvation Therapy Performance of Chlorin e6 Immobilized, Polydopamine-Coated Hollow, Porous Ceria-Based, Hypoxia-Tolerant Nanozymes Carrying a Cascade System

Microalgae show great promise for producing valuable molecules like biofuels, but their large-scale production faces challenges, with harvesting being particularly expensive due to their low concentration in water, necessitating extensive treatment. While methods such as centrifugation and filtration have been proposed, their efficiency and cost-effectiveness are limited. Flotation, involving air-bubbles lifting microalgae to the surface, offers a viable alternative, yet the repulsive interaction between bubbles and cells can hinder its effectiveness. Previous research from our group proposed using an amphiphilic chitosan derivative, polyoctyl chitosan (PO-chitosan), to functionalize bubbles used in dissolved air flotation (DAF). Molecular-scale studies performed using atomic force microscopy (AFM) revealed that PO-chitosan's efficiency correlates with cell surface properties, particularly hydrophobic ones, raising the question of whether this molecule can in fact be used more generally to harvest different microalgae. Evaluating this, we used a different strain of Chlorella vulgaris and first characterized its surface properties using AFM. Results showed that cells were hydrophilic but could still interact with PO-chitosan on bubble surfaces through a different mechanism based on specific interactions. Although force levels were low, flotation resulted in 84% separation, which could be explained by the presence of AOM (algal organic matter) that also interacts with functionalized bubbles, enhancing the overall separation. Finally, flocculation was also shown to be efficient and pH-independent, demonstrating the potential of PO-chitosan for harvesting microalgae with different cell surface properties and thus for further sustainable large-scale applications.

The Biophysical Properties of Microalgal Cell Surfaces Govern Their Interactions with an Amphiphilic Chitosan Derivative Used for Flocculation and Flotation

Superior flexibility and toughness can be achieved in bioactive hydrogels by the use of a double polymer network with complementary properties. Inspired by this design principle, we here combine polyacrylic acid (PAA) and sodium alginate (SA) to obtain a dual-reinforced double interpenetrating network (d-DIPN) hydrogel. The dual reinforcement involves ionic cross-linking and introduction of SiO2 nanoparticles, which leads to extraordinary improvements in strength and toughness. Compared with the standard PAA hydrogel that offers an elongation of 240% and a breakage stress of 0.03 MPa, the prepared SA(Ca2+)–PAA–SiO2 hydrogel shows an elongation above 1000% and a breakage stress of 1.62 MPa. Moreover, the combination of strong covalent cross-links and weak reversible interactions provides the d-DIPN hydrogel with swelling resistance and self-healing behavior, adhesive abilities, and shape memory performance. Furthermore, we show that the biocompatibility and bone cell proliferation ability of the hydrogels can be improved through a mineralization process despite an observed reduction in breakage strain and stress. Taken as a whole, our work paves the way for the design of strong and tough hydrogels, with potential applications within biomedicine and particularly tissue engineering.

Highly Stretchable, Swelling-Resistant, Self-Healed, and Biocompatible Dual-Reinforced Double Polymer Network Hydrogels

β-Galactosidase (β-Gala) is an essential biomarker enzyme for early detection of breast tumors and cellular senescence. Creating an accurate way to monitor β-Gala activity is critical for biological research and early cancer detection. This work used fluorometric, colorimetric, and paper-based color sensing approaches to determine β-Gala activity effectively. Via the sensing performance, the catalytic activity of β-Gala resulted in silicon nanoparticles (SiNPs), fluorescent indicators obtained via a one-pot hydrothermal process. As a standard enzymatic hydrolysis product of the substrate, kaempferol 3-O-β-d-galactopyranoside (KOβDG) caused the fluorometric signal to be attenuated on kaempferol-silicon nanoparticles (K-SiNPs). The sensing methods demonstrated a satisfactory linear response in sensing β-Gala and a low detection limit. The findings showed the low limit of detection (LOD) as 0.00057 and 0.098 U/mL for fluorometric and colorimetric, respectively. The designed probe was then used to evaluate the catalytic activity of β-Gala in yogurt and human serum, with recoveries ranging from 98.33 to 107.9%. The designed sensing approach was also applied to biological sample analysis. In contrast, breast cancer cells (MCF-7) were used as a model to test the in vitro toxicity and molecular fluorescence imaging potential of K-SiNPs. Hence, our fluorescent K-SiNPs can be used in the clinic to diagnose breast cellular carcinoma, since they can accurately measure the presence of invasive ductal carcinoma in serologic tests.

Triple Sensing Modes for Triggered β-Galactosidase Activity Assays Based on Kaempferol-Deduced Silicon Nanoparticles and Biological Imaging of MCF-7 Breast Cancer Cells

DNA has been demonstrated to be a versatile material for construction at the nanoscale. DNA nanodevices are highly programmable and allow functionalization with multiple entities such as imaging modalities (fluorophores), targeting entities (aptamers), drug conjugation (chemical linkers), and triggered release (photoresponsive molecules). These features enhance the use of DNA nanodevices in biological applications, catalyzing the rapid growth of this domain of research. In this review, we focus on recent progress in the development and use of aptamer-functionalized DNA nanodevices as theranostic agents, their characterization, applications as delivery platforms, and advantages. We provide a brief background on the development of aptamers and DNA nanodevices in biomedical applications, and we present specific applications of these entities in cancer diagnosis and therapeutics. We conclude with a perspective on the challenges and possible solutions for the clinical translation of aptamer-functionalized DNA nanodevices in the domain of cancer therapeutics.

Aptamer-Programmed DNA Nanodevices for Advanced, Targeted Cancer Theranostics

The growing threat of bacterial resistance is a critical global health concern, necessitating the development of more efficient methods for evaluating antimicrobial efficacy. Here, we introduce a technique based on the sensitivity of bacterial collective motion to environmental changes, using motion trajectory analysis for swift antibiotic susceptibility appraisal within a simple spread-out of bacterial droplet. By single cell tracking in bacterial fluids near the droplet edge or boundary-detection of the colony expansion, we achieved rapid evaluation of antibiotic efficacy in under 60 min. This method is not only faster than traditional assays but also provides insights into drug-bacterial interactions, offering a powerful tool for advancing both diagnostic testing and the development of antimicrobial agents.

Rapid Evaluation of Antimicrobial Potency Through Bacterial Collective Motion Analysis

As the core component of microbial fuel cells, the conductivity and biocompatibility of anode are hard to achieve simultaneously but significantly influence the power generation performance and the overall cost of microbial fuel cells. Stainless steel felt has a low price and high conductivity, making it a potential anode for the large-scale application of microbial fuel cells. However, its poor biocompatibility limits its application. This study provides a one-step binder-free modification method of a stainless steel felt anode with reduced graphene oxide to retain the high conductivity while greatly improving biocompatibility. The maximum power density achieved by reduced graphene oxide modified stainless steel felt was 951.89 mW/m2, 5.49 and 1.91 times higher than the unmodified stainless steel felt anode and reduced graphene oxide coated stainless steel felt by Nafion, respectively. The robust reduced graphene oxide modification markedly improved the biocompatibility by forming a uniform biofilm and utilizing the high conductivity of reduced graphene oxide to enhance the charge transfer rate. It led to 92.7 and 37.9% decreases in charge transfer resistance of reduced graphene oxide modified stainless steel felt compared to the unmodified one and the anode modified with reduced graphene oxide by Nafion, respectively. The excellent performance and green synthesis method of the anode validated its potential as a high-performance anode material for scaled-up microbial fuel cell applications.

COVID-19 has become one of the deadliest epidemics in the past years. In efforts to combat the deadly disease besides vaccines, drug therapies, and facemasks, significant focus has been on designing specific methods for the sensitive and accurate detection of SARS-CoV-2. Of these, surface-enhanced Raman scattering (SERS) is an attractive analytical tool for the identification of SARS-CoV-2. SERS is the phenomenon of enhancement of Raman intensity signals from molecular analytes anchored onto the surfaces of roughened plasmonic nanomaterials. This work gives an updated summary of plasmonic gold nanomaterials (AuNMs) and silver nanomaterials (AgNMs)-based SERS technologies to identify SARS-CoV-2. Due to extreme "hot spots" promoting higher electromagnetic fields on their surfaces, different shapes of AuNMs and AgNMs combined with Raman probes have been reviewed for enhancing Raman signals of probe molecules for quantifying the virus. It also reviews progress made recently in the design of certain specific Raman probe molecules capable of imparting characteristic SERS response/tags for SARS-CoV-2 detection.
