New 3-amino-5-ethenylcyclopentenones, myrothenones A (4) and B (5), were isolated together with known 6-n-pentyl-α-pyrone (1), trichodenone A (2), and cyclonerodiol (3) from the marine algicolous fungus of genus of Myrothecium. The structure and absolute stereochemistry of the new compounds were established by spectral interpretation and X-ray analysis. Compounds 1 and 4 exhibited a tyrosinase inhibitory activity with IC50 value of 0.8 and 6.6 μM, respectively, which are more active than kojic acid (IC50, 7.7 μM) currently being used as a functional personal-care compound.

Myrothenones A and B, Cyclopentenone Derivatives with Tyrosinase Inhibitory Activity from the Marine-Derived Fungus Myrothecium sp.

The multi-component fingerprinting method of Radix Salviae Miltiorrhizae (Root of Salvia miltiorrhiza BGE.), an important and popular medicinal herb in traditional Chinese medicine, was studied using reverse-phase HPLC and LC-MS-MS. Extract containing both the water-soluble phenolic compounds and nonpolar diterpenoid compounds known to be the herb's main bioactive components was prepared by a two-step extractive procedure. An HPLC fingerprinting method which can simultaneously separate these two types of compounds was established with gradient elution mode and photodiode array detection at 280 nm. Eighteen peaks in this HPLC fingerprint were structurally identified by employing LC-MS-MS techniques. The electrospray ionization (ESI) MS-MS spectra of most salvianolic acids displayed a characteristic behavior of loss of danshensu and caffeic acid moieties, while those of tanshinones showed a particular behavior of loss of H2O which is quite different from the fragmentation pattern in electron ionization mass spectrometry (EI-MS). The HPLC fingerprints of 7 batches of crude drugs showed similar separation pattern and provided much chemical information of the pharmacologically-active compounds in the crude drugs, which is useful for the authentication and quality evaluation of this medicinal herb.

Multi-component HPLC Fingerprinting of Radix Salviae Miltiorrhizae and Its LC-MS-MS Identification

From the MeOH extract of Vietnamese Caesalpinia sappan, a novel biogenetically exclusive benzindenopyran, with a new carbon framework, neoprotosappanin (1), and a new compound, protosappanin A dimethyl acetal (3), were isolated together with protosappanin E-2 (2), neosappanone A (4), and 13 previously reported phenolic compounds (5-17). Their structures were elucidated on the basis of spectroscopic data. Compounds 1-4, 7, 13, and 15-17 showed significant xanthine oxidase inhibitory activity in a concentration-dependent manner, and sappanchalcone (17) showed the most potent activity with an IC50 value of 3.9 microM, comparable to that of positive control allopurinol (IC50, 2.5 microM). The kinetic study of these inhibitors indicated that they are competitive inhibitors, the same as allopurinol, except for 1 and 16 which are noncompetitive inhibitors.

Xanthine Oxidase Inhibitors from the Heartwood of Vietnamese Caesalpinia sappan

New bisindole alkaloids, hyrtinadines C (1) and D (2), have been isolated from an Okinawan marine sponge Hyrtios sp. The structures of hyrtinadines C (1) and D (2) were elucidated based on analyses of the spectral data. Hyrtinadines C (1) and D (2) were the relatively rare alkaloids possessing a 3,4-fused azepinoindole skeleton. Hyrtinadines C (1) and D (2) showed antimicrobial activity.

Five new diketopiperazine alkaloids, brevicompanines D—H (3—7), together with two known analogs, allo-brevicompanine B (1) and fructigenine B (2), were isolated from a deep ocean sediment derived fungus Penicillium sp. Their structures were established by spectroscopic methods including 2D NMR and chiral HPLC analysis. Compounds 4 and 7 inhibited lipopolysaccharide (LPS)-induced nitric oxide production in BV2 microglial cells.

Diketopiperazine Alkaloids from a Deep Ocean Sediment Derived Fungus Penicillium sp.

We have investigated the base-induced long-range halogen dance reactions of 4,5-dibromo- or 4-bromo-5-iodothiazoles bearing sulfur-containing aromatic heterocycles at the C2-position. We have found that the reaction occurs in bithiazole regioisomers or (thiophenyl)thiazole derivatives, in which the C-5 halo group on the thiazole halogen donor regioselectively migrates to a halogen acceptor ring after treatment with lithium bis(trimethylsilyl)amide. The substrate with a thiophen-2-yl substituent required highly basic P4-t-Bu to induce the halogen dance reaction.

Long-Range Halogen Dance Reaction in 4,5-Dihalogeno-2-(Sulfur-Containing Heterocyclyl)thiazole

The main ingredients of Maobushisaishinto (MBST) are ephedrine (EP), methyl eugenol (ME), and aconitine (AC). The pharmacological effects are presumed to be due to the combined effects of these ingredients. In this study, we investigated the impact of the particles present in MBST suspensions on the absorption of the ingredients. Coarse, colloidal, and molecular dispersions were detected when MBST was dispersed in water at 25 and 70 °C. Regardless of temperature, the ratio of MBST in molecular dispersions was the highest, and the ratio of coarse dispersions was greater than that of colloidal dispersions. Particles ranging from 50 to 900 nm were observed in the colloidal dispersions prepared by treatment at 25 °C for 3 min. However, in 70 °C water, the mean particle size decreased, and the number of nanoparticles tended to increase. The levels of EP, ME, and AC in molecular dispersions were higher than those in coarse and colloidal dispersions, with no significant difference observed between the coarse and colloidal dispersions. On the other hand, in small intestinal penetration, the levels of EP, ME, and AC in colloidal dispersions were higher than those in the other two dispersions. Moreover, adding colloidal particles to the dissolved drug (molecular dispersions) increased the drug's permeability through the small intestinal membrane. In conclusion, colloidal particles are produced when MBST is suspended. Furthermore, we showed that these colloidal particles enhance the absorption of the main ingredients of MBST.

Colloidal Particle in Suspensions of Maobushisaishinto Extract Granules Enhances Drug Intestinal Penetration

A total synthesis of javaberine A was achieved through a lithium amide-mediated intramolecular hydroamination of an N-allyl aminoalkene. The desired hydroamination was accomplished using an excess of i-Pr2NH with a substoichiometric amount of n-BuLi. Using an excess of both n-BuLi and i-Pr2NH led to tandem cyclization, however, resulting in the construction of a tricyclic structure through the formation of one C–N and two C–C bonds in a single operation. Additionally, epimerization of the H8-H14 cis-benzyl tetrahydroisoquinoline to the trans isomer was achieved via β-elimination followed by intramolecular hydroamination.

Synthetic Studies of Javaberine A Based on Intramolecular Hydroamination of Alkenes

Pharmaceutical packaging is essential for enhancing the storage stability of medicine and can improve medication adherence and usability. Despite their widespread use, blister packs can be challenging to use, especially when pushing out the medication. This study investigates how specific cavity characteristics of blister packs can enhance usability for spherical capsules, which are harder to push out than tablets. The findings of this study show that reducing the thickness of the unformed sheet, or the thickness at the top and corners of the cavities, reduces the effort required to push out the capsules. Similarly, for cavities with different shapes, reducing the thickness at the top and increasing the corner radius also eases the push-out process. These insights emphasize the importance of systematic design in pharmaceutical packaging to improve patient medication adherence.
