Advanced Synthesis and Catalysis

Engineered myoglobins have recently emerged as promising scaffolds for catalyzing carbene-mediated transformations. In this work, we investigated the effect of altering the metal center and its first-sphere coordination environment on the carbene transfer reactivity of myoglobin. To this end, we first established an efficient protocol for the recombinant expression of myoglobin variants incorporating metalloporphyrins with non-native metals, including second- and third-row transition metals (ruthenium, rhodium, iridium). Characterization of the cofactor-substituted myoglobin variants across three different carbene transfer reactions (cyclopropanation, N-H insertion, S-H insertion) revealed a major influence of the nature of metal center, its oxidation state and first-sphere coordination environment on the catalytic activity, stereoselectivity, and/or oxygen tolerance of these artificial metalloenzymes. In addition, myoglobin variants incorporating manganese- or cobalt-porphyrins were found capable of catalyzing an intermolecular carbene C-H insertion reaction involving phthalan and ethyl α-diazoacetate, a reaction not supported by iron-based myoglobins and previously accessed only using iridium-based (bio)catalysts. These studies demonstrate how modification of the metalloporphyrin cofactor environment provides a viable and promising strategy to enhance the catalytic properties and extend the reaction scope of myoglobin-based carbene transfer catalysts.

Abstract The control of selectivity in the reactions of the highly reactive open‐shell carbon radicals is an attractive but often challenging task. Building on the strategy of photoinduced iminyl radical‐mediated C−C bond cleavage, we have developed photocatalytic neophyl rearrangement and reduction of distal carbon radicals under visible light irradiation of O ‐acyl oximes. This mild protocol tolerates a wide range of readily available O ‐acyl oximes, enabling facile synthesis of diversely substituted α,β ‐unsaturated nitriles and β ‐functionalized saturated nitriles in a highly selective manner. magnified image

Photocatalytic Neophyl Rearrangement and Reduction of Distal Carbon Radicals by Iminyl Radical‐Mediated C−C Bond Cleavage

Abstract The synthesis of aryl amines via the formation of a C−N bond is an essential tool for the preparation of functional materials, active pharmaceutical ingredients and bioactive products. Usually, this chemical connection is only possible by transition metal‐catalyzed reactions, photochemistry or electrochemistry. Here, we report a metal‐free arene C−H amination using hydroxylamine derivatives under benign conditions. A charge transfer interaction between the aminating reagents TsONHR and the arene substrates enables the chemoselective amination of the arene, even in the presence of various functional groups. Oxygen was crucial for an effective conversion and its accelerating role for the electron transfer step was proven experimentally. In addition, this was rationalized by a theoretical study which indicated the involvement of a dioxygen‐bridged complex with a “Sandwich‐like” arrangement of the aromatic starting materials and the aminating agents at the dioxygen molecule. magnified image

A Metal‐Free Direct Arene C−H Amination

Abstract Itaconic acid is a chemically versatile unsaturated diacid that can be produced by fermentation and potentially it can replace petrol‐based monomers such as maleic and fumaric acids in the production of curable polyesters or new biocompatible functionalized materials. Unfortunately, due to the presence of the unsaturated CC bond, polycondensation of itaconic acid is hampered by cross reactivity and isomerization. Therefore, enzymatic polycondensations would respond to the need of mild and selective synthetic routes for the production of novel bio‐based polymers. The present work analyses the feasibility of enzymatic polycondensation of diethyl itaconate and, for the first time, provides comprehensive solutions embracing both the formulation of the biocatalyst, the reaction conditions and the choice of the co‐monomers. Computational docking was used to disclose the structural factors responsible for the low reactivity of dimethyl itaconate and to identify possible solutions. Surprisingly, experimental and computational analyses revealed that 1,4‐butanediol is an unsuitable co‐monomer for the polycondensation of dimethyl itaconate whereas the cyclic and rigid 1,4‐cyclohexanedimethanol promotes the elongation of the oligomers. magnified image

Understanding Potentials and Restrictions of Solvent‐Free Enzymatic Polycondensation of Itaconic Acid: An Experimental and Computational Analysis

IIn this paper, a visible‐light‐induced intramolecular [2+2] cycloaddition has been revealed to synthesize the bishomocubanone derivatives in moderate to excellent yield under mild conditions. This method demonstrated a broad substrate scope and excellent functional‐group tolerance, affording cubane precursors and a series of bishomocubanones. Meanwhile, a hundred‐gram scale experiment of this method has been employed to synthesize cubane precursors with 98% yield.

Here, we present an innovative approach to achieve Minisci‐type alkylation of cyclic N‐ketimines, enabling access to a diverse range of valuable alkylated sulfonyl ketimines via iron hydride hydrogen atom transfer. More than 35 examples are shown with a wide range of substrates. Additionally, this system is easy to operate and can be efficiently scaled up. Mechanistic investigations revealed that the alkyl radical process involved and the air acts as the sole oxidant in this system.

Iron Catalyzed Radical Alkylation of Cyclic Ketimines with olefins via Metal Hydride Atom Transfer

A half‐sandwich nickel complex containing an NHC‐carboxylate chelate ligand (i.e., [Ni(η5‐Cp)(κ2‐C,O‐NHC)]) derived from the (S)‐leucine has been identified for the selective hydrothiolation of terminal acetylenes toward challenging Markovnikov‐type 𝛼 vinyl thioethers, even with aryl alkynes. The activity and selectivity exhibited by the nickel complex are comparable, if not superior, to those of complexes of rhodium reported to be the best catalysts to date for the Markovnikov addition in the thiol‐yne reaction. The experimental data are consistent with the participation of the coordinated carboxylate group in the catalytic reaction as an intermolecular acid‐base co‐catalyst, favoring the formation of nickel‐thiolate intermediates.

A copper and DMAP relay‐catalyzed strategy has been achieved for the modular synthesis of 2‐alkoxy quinolines from a three‐component coupling of terminal alkynes, anthranils, and alcohols. In this protocol, a novel quinoline pyridine salt is formed and serves as a key intermediate in the formation of 2‐alkoxy quinolines. The reaction has a broad substrate scope of each reaction partners, affording diversified 2‐alkoxy quinoline products in generally good yields.

Herein, we report regiodivergent nucleophilic attack of Grignard reagents to 2‐alkynyl aziridines. Under the catalysis of a Cu(I)‐diphosphine complex, benzylic Grignard reagents add to the carbon‐carbon triple bond of the aziridines to deliver α‐allenylamines in moderate to excellent yields; the direct addition of the Grignard reagents to the aziridines in the absence of catalyst provided homopropargylic amine derivatives. Both reactions could proceed in highly enantiospecific ways. Further synthetic manipulations of these products enable rapid access to diverse useful heterocyclic molecular scaffolds.
